不同提取方法对佛手多糖性质和乙醇脱氢酶活性的影响

摘要:目的:研究不同提取方法对佛手多糖理化性质及乙醇脱氢酶活性的影响,为改进佛手多糖生产方式和佛手多糖解酒研究提供依据。方法:分别采用传统水提法、超声辅助法、微波辅助法、超声-微波协同提取法和复合酶法提取佛手多糖,探讨五种提取方法对多糖得率、糖醛酸含量、分子量、单糖组成、红外光谱和乙醇脱氢酶活性的影响。结果:水提法和超声-微波协同提取多糖得率最高,分别为4.26±0.21%和4.15±0.14%。不同方法提取的佛手多糖分子量大小存在差异,其中复合酶法提取多糖平均分子量为617.85 KD,约是水提多糖分子量的4倍。五种多糖均呈现出典型的吡喃型多糖和α型糖苷键吸收,单糖组成均以阿拉伯糖、半乳糖、葡萄糖为主。超声-微波协同提取多糖对乙醇脱氢酶激活能力最强,可达46.58±1.76%,而传统水提多糖抑制乙醇脱氢酶活性。结论:超声-微波协同提取佛手多糖得率高且所需时间短,对乙醇脱氢酶激活能力强,推测乙醇脱氢酶激活能力与多糖分子量大小、半乳糖醛酸连接方式有关。

Effect of different extraction methods on characteristics and alcohol dehydrogenase activity of finger citron polysaccharides
Abstract:Objective: The effect of different extraction methods on characteristics and alcohol dehydrogenase (ADH) activity of finger citron polysaccharides (FCPs) were investigated to provide the references for the development of extraction methods and hepatoprotective activity of FCPs. Method: FCPs were extracted by five extraction methods, including traditional water extraction(TWE), ultrasonic assisted extraction(UE), microwave assisted extraction (ME), ultrasonic-microwave synergistic extraction (UMSE) and compound enzyme assisted extraction (CEE). The impacts of different extraction methods on the yield of polysaccharides, uronic acid content, molecular weight, monosaccharide composition, IR spectrum and ADH activity were investigated. Result: The results showed that the yields of FCPs extracted by TWE and UMSE were 4.26±0.21% and 4.15±0.14%, respectively, which are higher than other extraction methods. There were differences in molecular weight of FCPs prepared by different extraction methods. Average molecular weight of polysaccharides prepared by CEE was 617.85KDa, about 4 times than the polysaccharides by TWE. Five polysaccharides had typical absorption for pyran polysaccharides andα-glycosidic bond. Monosaccharide composition mainly included arabinose, galactose, glucose. The polysaccharides extracted by UMSE had the highest activating effect on ADH, with a pencentage activation of 46.58±0.26%, but polysaccharides extracted by TWE inhibited the activity of ADH. Conclusion: The polysaccharide prepared by UMSE had good yield, high efficiency and highest activating effect on ADH. The activation of ADH may be due to the molecular weightandgalacononic acid connection.
