邻苯二甲酸二(2-乙基己基)酯对不同品系雄性大鼠肝脏毒性及生殖毒性的研究

摘要:【目的】分析常见不同品系大鼠对邻苯二甲酸二(2-乙基己基)酯(DEHP)肝脏毒性以及雄性生殖毒性的敏感度,为DEHP毒性研究探索合适的动物模型。【方法】选择常见的SD和Wistar大鼠,在其青春期进行维持4周的不同剂量的DEHP染毒处理。观察染毒后体重和各脏器的变化,测定两种品系大鼠在不同的染毒剂量下血清中睾酮水平、碱性磷酸酶(ALP)、谷丙转氨酶(ALT)、谷草转氨酶(AST)含量和肝脏中MDA水平,并通过病理切片观察组织损伤变化。【结果】经DEHP暴露后,SD大鼠的体重和脏器质量较对照组出现了显著变化,而Wistar大鼠的脏器重量在染毒后未出现异样。DEHP对两种大鼠脾脏以及肾脏的重量均无显著影响。在SD大鼠中,低剂量DEHP处理使血清中ALP、ALT和AST的含量有下降趋势,随着DEHP剂量的升高,高剂量组血清中ALP、ALT和AST的含量则显著高于对照组;Wistar大鼠则在较低剂量的DEHP染毒情况下即发现ALP和ALT的升高,但高剂量处理时则无此现象。组织病理学分析,低剂量DEHP处理未对两种大鼠的肝脏造成明显损伤,但中、高剂量DEHP处理使SD大鼠肝细胞轻微脂肪变性同时使Wistar大鼠肝脏组织出现轻微小叶中心性肝细胞脂肪变性。此外,SD大鼠的血清睾酮含量随DEHP染毒剂量增加逐渐降低,Wistar大鼠的睾酮含量未随DEHP染毒剂量变化而变化。【结论】针对DEHP的肝脏毒性和雄性生殖毒性,SD大鼠表现出更明显的肝脏毒性和雄性生殖毒性敏感性。

Study on the liver toxicity and reproductive toxicity on male rats by di(2-ethylhexyl) phthalate
Abstract:【Objective】In this study, the susceptibility of differentspecies of rat to the liver toxicityand male reproductive toxicity caused by DEHP was analyzed to find out an appropriate animal model for studiesonDEHP toxicity.【Methods】SD and Wistar rats were treated with different doses of DEHP for four weeksduring puberty. The changes of body weight and organs index were observed. The levels of testosterone, ALP, ALT, AST in serum and MDA in liver were measured. Histological changes of liver were also observed. 【Results】After DEHP exposure, body weight and organ quality of SD rats changed significantly compared with control group, while body weight and organ in Wistar rats did not. DEHP had no significant effects on spleen weight and kidney weight in both kinds of rats. In SD rats, there were slight decrease of levels of ALP, ALT and AST in serum during lower dose DEHP exposure, whilst elevated significantly during high dose DEHP exposure.Wistar rats showed elevated level of ALP and ALT during low doses DEHP exposure,but not during high dose exposure. Pathological analysis showed that low doses of DEHP did not cause significant damage to the liver of both kinds of rats, while moderate and high doses of DEHP caused mild fatty degeneration in hepatocytes of rats. Besides, the level of serum testosterone decreased significantly in SD rats but not in Wistar rats. 【Conclusion】SD rats showed more obvious susceptibility to liver toxicity and male reproductive toxicity than that of Wistar ratsto DEHP exposure.
