改善结直肠癌的膳食因子的筛选及其诱导细胞凋亡作用的研究

摘要:目的:筛选改善结直肠癌的膳食因子,并对其诱导细胞凋亡作用进行研究。方法:采用噻唑蓝法检测16种膳食因子醇提物对不同结肠癌细胞抑制作用;建立荷瘤小鼠模型初筛能够改善结肠癌作用的膳食因子;采用蛋白质印迹法测定黑胡椒醇提物对结肠癌细胞凋亡的影响。结果:黑胡椒和丁香的乙醇提取物具有极其显著的癌细胞增殖抑制作用;黑胡椒、丁香和薏苡仁的膳食干预对荷瘤小鼠具有显著的抑制肿瘤、延长生存期的作用,丁香和薏苡仁可改善荷瘤小鼠的抓力;黑胡椒醇提物能够上调结肠癌细胞的Bad、LC3B表达水平,下调Bcl-2、Akt和磷酸化Akt表达水平,从而促进癌细胞凋亡。结论:黑胡椒、丁香、薏苡仁能够改善结肠癌,以黑胡椒的综合改善效果最佳;黑胡椒可能通过诱导细胞凋亡途径改善结肠癌及其恶病质。

Screening of dietary factors to improve colorectal cancer and its induction of apoptosis
Abstract:Objective: To screen dietary factors that could improve colorectal cancer and to study its effects on inducing apoptosis. Methods: The inhibitory effect of alcohol extracts of 16 dietary factors on different colon cancer cells was detected by thiazolyl blue method; dietary factors that can improve colon cancer function were established by the tumor-bearing mouse model; and the alcohol extracts of black pepper were determined by Western blotting. The effect of apoptosis in colon cancer cells. Results: The study found that the ethanol extracts of black pepper and clove have extremely significant inhibitory effects on the proliferation of cancer cells; the dietary interventions of black pepper, clove and coix seed have significant effects on inhibiting tumor volume and prolonging survival time in tumor-bearing mice. Dietary intervention with clove and coix seed can improve the general quality of life of tumor-bearing mice; Black pepper extract can up-regulate the expression of Bad and LC3B in colon cancer cells and down-regulate Bcl-2, Akt, and phosphorylated Akt expression levels. , which promotes apoptosis of cancer cells. Conclusion: Black pepper, clove and coix seed can improve the colorectal cancer and its cachexia, and the black pepper has the best comprehensive improvement effect. Black pepper may improve the colon cancer and its cachexia by inducing apoptosis pathway.
