离子交换树脂吸附法分离叔亮氨酸

摘要:葡萄糖脱氢酶辅酶再生过程中会产生高浓度葡萄糖酸,葡萄糖酸与氨基酸具有相似的物理化学性质,分离困难,限制了酶法不对称还原制备手性氨基酸的工业应用。本论文针对酶促不对称还原三甲基丙酮酸生产叔亮氨酸工艺,采用离子交换树脂吸附法,首先通过静态吸附实验筛选和优化了吸附条件,发现凝胶类树脂在低pH条件下有利于提高叔亮氨酸吸附率和吸附量。进一步优化动态吸附条件,采用多柱串联工艺,有效提高了叔亮氨酸吸附率和吸附量,分别达到96.5%和0.97mmol/g。研究结果在2吨工业生产规模进行放大,叔亮氨酸收率达到88.1%,化学纯度达到99%。

Extraction of L-tert-leucine by ion exchange resin adsorption
Abstract:High concentration of gluconic acid will be produced during the regenerationofcoenzyme by glucose dehydrogenase .Gluconic acid and amino acid have similar physical and chemical properties, and the separation is difficult,which restricts the industrial application of asymmetric reduction of chiral amino acids by enzymatic method.For production of L-tert-leucine by enzymatic asymmetric reduction of three methyl pyruvic acid,ion exchange resin adsorption method is adopted in this paper.At first,the screening conditions were screened and optimized by static adsorption experiments. it was found that the gel resin is beneficial to improve the adsorption and adsorption of L-tert-leucine under low pH conditions.Further optimizing the dynamic adsorption conditions,by using multi column series technology, the adsorption rate and adsorption rate of L-tert-leucine are improved, and 96.5% and 0.97mmol/g respectively.The results were amplified in the 2 tons industrial scale, and the yield of L-tert-leucine reaches 88.1% and the chemical purity reaches 99%.
