摘要：目的：研究全反式维甲酸（ATRA）与苦丁冬青（Ilex kudingcha C.J.Tseng，KD）联合作用对乳腺癌的生长抑制效果。方法：采用FVB/N-Tg（MMTV-PyVT）转基因自发乳腺癌小鼠模型，给予不同药物，给药28天后眼球取血法取血检测血脂四项，处死小鼠后剥离肿瘤组织拍照并测量重量；采用免疫组化方法检测肿瘤组织中增殖活性标志蛋白Ki67的表达水平，用蛋白质免疫印迹法检测增殖相关蛋白Cyclin D1、p21，Caspase细胞凋亡通路主要蛋白水平。结果：（1）ATRA对小鼠TC、TG有一定的升高作用，而联合苦丁冬青后可以改善高血脂水平，联合能显著降低低密度脂蛋白含量，而高密度脂蛋白显著升高；（2）联合能显著抑制肿瘤生长，与其他三种处理相比肿瘤体积明显减小、重量显著降低；（3）联合处理使得肿瘤组织中Ki67阳性细胞数目显著减少，Cyclin D1蛋白表达量显著降低，但p21无显著变化；（4）联合处理显著增加Cleaved Caspase9蛋白表达量并减少Caspase7表达量。结论：ATRA联合苦丁冬青能抑制FVB/N-Tg（MMTV-PyVT）转基因小鼠乳腺肿瘤生长，可能依赖于Caspase通路诱导细胞凋亡。
The inhibitory effect of ATRA combined with Ilex kudingcha C.J.Tseng on breast cancer

Abstract： Objective: To investigate the inhibitory effects of ATRA combined with Ilex kudingcha C.J.Tseng (KD) on the growth of breast cancer. Methods: FVB/N-Tg (MMTV-PyVT) transgenic spontaneous breast cancer mice were used as study model with different drug treatments. After 28 days of administration, blood samples were taken from the eyes for examining the levels of serum TG, TC, HDL-c and LDL-c. The mice were sacrificed and dissected the tumor. The Ki67 protein level was assessed with immunohistochemical. The proliferative related-protein Cyclin D1, p21 and the apoptosis related-protein Caspase were examined by immunoblotting. Results: (1) The levels of TC and TG were significantly increased by ATRA, ATRA combined with Ilex kudingcha C.J.Tseng can ameliorated the TC and TG levels. LDL-c was significant decreased and HDL-c significant increased in the combination treated group. (2) The tumor growth was significantly inhibited, the tumor volume and weight were significantly reduced in the combination treated group; (3) The number of Ki67 positive cells and the expression of Cyclin D1 were signficantly decreased, but the expression of p21 was not affected in the combination treated group; (4) The expression of Cleaved Caspase9 was significantly increased and Caspase7 was decreased in the combination treated group. Conclusion: ATRA combined with Ilex kudingcha C.J.Tseng inhibit the growth of mammary tumor in FVB /N-Tg (MMTV-PyVT) transgenic mice, likely via the Caspase pathway to induce cell apoptosis.
