小叶苦丁茶粗提物与雷帕霉素联合通过阻滞细胞周期抑制乳腺癌细胞增殖

摘要:目的:探究小叶苦丁茶粗提物与雷帕霉素联合对乳腺癌细胞增殖、周期和凋亡的影响。方法:小叶苦丁茶粗提物和雷帕霉素单独及联合分别处理HCC1806和MDA-MB-231两种乳腺癌细胞,先采用显微镜观察、血球计数法、CCK8细胞增殖实验检测两种物质单独及联合对细胞生长、增殖的影响;接着采用流式细胞术和蛋白质免疫印迹法检测细胞周期和凋亡及相关蛋白质的表达水平。结果:在两种细胞类型中,相比于对照组和单独处理组,联合组,(1)活细胞数目明显减少;(2)细胞存活率降低;(3)G0/G1期细胞所占比例升高,p21蛋白质的表达量增加,而p27和CyclinD1无明显变化;(4)细胞凋亡率和凋亡相关蛋白(PARP和Bcl-2)均无明显差异。结论:小叶苦丁茶粗提物和雷帕霉素联合可以抑制HCC1806和MDA-MB-231两种乳腺癌细胞增殖,并通过上调p21导致细胞G1期阻滞。

Combination of Ligustrum robustum extracts with rapamycin inhibits breast cancer cells proliferation via arresting cell cycle
Abstract:Objective: To investigate the cell proliferation, cell cycle and apoptosis of Ligustrum robustum extracts combined with rapamycin on breast cancer cells. Methods: HCC1806 and MDA-MB-231 were treated with Ligustrum robustum extracts and rapamycin or both, and then cell growth was assessed by viable cell counting assay, cell cycle and apoptosis were examined by flow cytometer analysis and related proteins were determined by immunoblotting. Results showed that combination of Ligustrum robustum extracts and rapamycin significantly reduced the number of viable cells and cell survival rate, increased the proportion of cells in G0/G1 phrase and the expression of p21, not p27, cyclinD1, PARP and Bcl-2. Conclusion: Combination of Ligustrum robustum extracts with rapamycin inhibits HCC1806 and MDA-MB-231 cells proliferation via arrestring cell cycle.
