TiO2纳米材料对前成骨细胞MC3T3-E1增殖分化及免疫毒性研究

摘要:通过研究不同晶型、浓度TiO2对成骨细胞的增殖和分化能力及免疫毒性的影响。方法:CCK-8法检测细胞增殖能力、定量测定碱性磷酸酶(ALP)含量、酶联免疫吸附(ELISA法)测定IL-1、IL-6水平。结果:纳米TiO2材料对细胞增殖存在抑制作用,不同晶型材料毒性不同。对MC3T3-E1分化抑制效果同样存在差异,当纳米颗粒浓度较低时,锐钛矿材料对细胞分化抑制效果较强;纳米颗粒浓度增大时,金红石材料对细胞分化抑制效果提高。免疫因子IL-1分泌与纳米颗粒晶型关联较小,而IL-6与纳米颗粒晶型有较大关系,金红石材料刺激MC3T3-E1分泌更多IL-6。相同浓度下,培养时间短时,IL-1分泌水平较高,随时间增加,与金红石材料共培养的细胞培养上清液中IL-6水平下降尤为明显。结论:纳米TiO2材料作为人工植入物材料之一,其细胞毒性不可忽视,其毒性与材料晶型、浓度、时间均存在一定关系。

Effect of nano-TiO2 with different characters on bone formation and immunotoxicity in MC3T3-E1
Abstract:Object: To evaluate the effect of nano-TiO2 with various physical characters on cell growth, differentiation and immunotoxicity. Methods: Using CCK-8 assay detect the cell proliferation, measuring alkaline phosphatase(ALP) content quantitatively, using enzyme-linked immunosorbent assay(ELISA) measured IL-1,IL-6 content. Result: Nano-TiO2 has inhibitary effect on cell proliferation, and it makes difference with different crystalization. The differentiation effect on MC3T3-E1 also has difference, when the concentration of nanoparticles is low, anatase materials inhibited more. When the concentration of nanoparticles increases, the inhitition effect of rutile materials increases. Nanoparticle crystalization has little connection with IL-1, while it makes difference with IL-6, rutile materials stimulate more serection of IL-6. In the same concentration, the concentration ofIL-1 is in a high level in a shorter incubation time. When the incubation time increases, the concentration of IL-6 in cell culture supernate which cocultured with rutile materials decreases obviously. Conclusions: As one of the artificial implants materials, the cytotoxicity of Nano-TiO2 matarials can\'t be ignored, which is related to crystalization, concentration and time.
