新型有机硒化合物乙烷硒啉对人舌鳞状细胞癌细胞的增殖、周期、凋亡及迁移的影响

摘要:目的:通过体外细胞实验探讨新型有机硒化合物乙烷硒啉(BBSKE)对人舌癌CAL27细胞株增殖活性、细胞周期、细胞凋亡及细胞迁移的影响。方法:采用MTT检测不同浓度BBSKE对CAL27细胞增殖的抑制作用,采用 Hoechst 33258免疫荧光染色检测不同浓度BBSKE对CAL27细胞凋亡的改变,采用流式细胞术检测不同浓度BBSKE对CAL27细胞的凋亡率和细胞周期分布的影响,采用Transwell小室检测不同浓度BBSKE对CAL27细胞迁移的影响。结果:BBSKE对CAL27细胞的生长增殖有显著的抑制作用(P<0.01),且具有药物浓度依赖性和时间依赖性。通过Hoechst 33258免疫荧光染色,BBSKE能够明显诱导CAL27细胞凋亡和凋亡形态改变。Annexin V-FITC/PI双染流式细胞术结果显示与对照组相比,BBSKE可使CAL27细胞凋亡率明显增加(P<0.01),并引起细胞周期阻滞于G2/M期(P<0.01)。Transwell小室实验结果显示,CAL27细胞的迁移细胞数随BBSKE浓度的升高而减少,BBSKE对CAL27细胞迁移有明显的抑制作用(P<0.01)。结论:BBSKE对CAL27细胞具有显著的增殖抑制作用,其通过引起细胞周期阻滞于G2/M期诱导细胞凋亡,并且抑制细胞迁移,提示BBSKE可作为TSCC患者化疗治疗的备选药物,本研究为其临床的应用提供实验依据。

Effects of ethaselen, a novel organoselenium compound, on proliferation, cycle, apoptosis and migration in human tongue squamous cell carcinoma cells.
Abstract: Objective: To investigate the effects of novel organic selenium compound Ethaselen (BBSKE) on the proliferation activity, cell cycle, apoptosis and cell migration of human tongue cancer CAL27 cell line in vitro. Methods: MTT was used to detect the inhibitory effect of different concentrations of BBSKE on the proliferation of CAL27 cells. The apoptosis of CAL27 cells was detected by Hoechst 33258 immunofluorescence staining. Results: BBSKE could significantly inhibit the growth and proliferation of CAL27 cells (P <0.01), and had drug concentration-dependent and time-dependent effects. By Hoechst 33258 immunofluorescence staining, BBSKE could obviously induce the apoptosis and morphological changes of CAL27 cells. Annexin V-FITC / PI staining showed that compared with the control group, BBSKE could significantly increase the apoptosis rate of CAL27 cells (P <0.01) and cause cell cycle arrest at G2 / M phase (P < 0.01). The results of Transwell chamber showed that the number of migrated cells in CAL27 cells decreased with the increase of BBSKE concentration, while BBSKE significantly inhibited the migration of CAL27 cells (P <0.01). Conclusions:BBSKE can significantly inhibit the proliferation of CAL27 cells. BBSKE can induce cell cycle arrest in G2 / M phase and inhibit cell migration, suggesting that BBSKE can be used as an alternative chemotherapy for chemotherapy in TSCC patients.This study provides the experimental basis for its clinical application.
