转录因子AP-2α表达下调导致斑马鱼胚胎脑出血

摘要:目的:验证斑马鱼胚胎脑血管发育是否受到转录因子AP-2α基因表达调控。方法:利用吗啉修饰的反义寡核苷酸(morpholino antisense oligos, MO)下调tfap-2α基因的表达;通过共聚焦显微镜观察了斑马鱼脑出血和血脑屏障(Blood-brain barrier, BBB)的渗漏情况。结果:研究发现转录因子AP-2α基因表达下调的斑马鱼胚胎呈现脑出血现象。进一步的研究证实转录因子AP-2α下调诱导了BBB的通透性增加。结论:转录因子AP-2α与斑马鱼胚胎脑血管的正常发育有关。

Blocking AP-2α Caused Cerebral Hemorrhage in Zebrafish Embryos
Abstract:Objective: To examine if cerebral vascular development in zebrafish embryos is regulated by transcription factor AP-2α. Methods: By using Morpholine antisense oligos (MO) to knockdown the expression of tfap-2α. Zebrafish cerebral hemorrhage and BBB leakage were observed by confocal microscopy.Results: In present study, its was that the expression of the transcription factor AP-2α was down-regulated in zebrafish embryos with cerebral hemorrhage. Further studies confirmed that down-regulation of the transcription factor AP-2α induces increased permeability of the BBB.Conclusion:The transcription factor AP-2α is associated with the normal development of zebrafish embryonic cerebral blood vessels.
