正交设计优选大黄甘草汤的提取工艺

摘要:目的:研究大黄甘草汤水提工艺的影响因素,确定最佳的制备工艺。方法:采用紫外分光光度法测定大黄总蒽醌类和甘草总黄酮类提取量,以大黄素和芦丁做对照品,显色剂分别是醋酸镁甲醇溶液和10%KOH溶液,检测波长依次为440nm和410nm。以大黄总蒽醌类、甘草总黄酮类提取量和干膏率的综合评分为指标,通过正交试验对浸泡时间、提取时间、粉碎度和料液比对大黄甘草汤水提工艺的影响进行考察。结果:最佳水提取工艺为粉碎度为0,浸泡0h,加12倍量水提取,每次1h。结论:通过正交设计优选的提取工艺简单、经济、可行,为大黄甘草汤的临床应用提供参考。

Optimization of extraction process of Rhei and Glycyrhizae Decoction by orthogonal test
Abstract:Objective: To study the factors of extraction process of Rhei and Glycyrhizae Decoction, optimizing the perfect one. Methods: UV-2300 was employed and detection wavelengths of 440,410nm with agents of emodin and rutinum, for achieving the determination of total rhubarb anthraquinones and total licorice flavonoids extraction.With contents of rhubarb anthraquinone, licorice flavonoidsand extraction rate as comprehensive evaluation indexes,orthogonal test was adopted to explore the effects of the soaking time, extracting time, crushing and solid-liquid ratio on water extracting technology. Results:The optimum water extraction process was: extracted twice with 12 times the amount of water for 1h per time after soaked in water for 0h. Conclusion:These optimized technologies are simple, economic and feasible, providing reference for clinical application of Rhei and Glycyrhizae Decoction.
