基于细胞膜片技术构建组织工程真皮:骨髓间充质干细胞、内皮细胞、成纤维细胞联合培养最适比例的研究

摘要:目的:研究骨髓间充质干细胞、脐静脉内皮细胞、真皮成纤维细胞在何种比例下联合培养可以形成组织工程真皮,为未来组织工程真皮的构建提供新思路。方法:HUVECs和HDF的复苏、培养,rBMSCs分离提取,以及共培养构建细胞膜片;1、3、7、14天细胞膜片HE染色以及厚度的测量、CD31免疫荧光染色、Ⅰ型、Ⅲ型胶原的免疫组化染色;第7天细胞膜片中的骨髓间充质干细胞干性的鉴定。结果:原代HUVECs、HDF复苏后形态良好,增殖旺盛;rBMSCs呈均一的长梭形,增殖旺盛;显微镜下观察第7天时成纤维细胞60%和80%组、第14天时40%和60%组细胞排列杂乱,编织成致密的网络状;CD31免疫荧光显示第7天、第14天时成纤维细胞60%、80%组内皮细胞形成的管腔状结构最为典型;免疫组化染色显示在第7天时成纤维细胞60%、80%组最接近人类真皮Ⅰ型胶原的含量,在第1天和第7天时成纤维细胞60%、80%组最接近人类真皮Ⅲ型胶原的含量;流式细胞仪鉴定显示在7天的各组细胞膜片中均有CD90标记的干细胞出现。结论:兔骨髓间充质干细胞、人脐静脉内皮细胞和人真皮成纤维细胞以9:5:21(即成纤维细胞60%组)的比例联合培养形成的细胞膜片在培养第7天时具有真皮的部分属性。

Construction of tissue engineering dermis based on cell membrane technique:the study of the optimal proportion of bone marrow mesenchymal stem cells,endothelial cells and fibroblasts
Abstract:The proportion of co culture of bone marrow mesenchymal stem cells(BMSCs), umbilical vein endothelial cells (HUVECs)and dermal fibroblasts(HDFs) can form tissue engineering dermis, which is helpful for the construction of tissue engineering dermis in the future. Method:Recoverd and cultured HUVECs and HDF ,separation and extraction of rBMSCs,Co-culture and construction of cell sheets,HE staining and Measurement of thickness of cell sheets on 1, 3, 7, 14 days,immunofluorescence staining of CD31,Immunohistochemical staining of type I and type III collagen,Identification of bone marrow mesenchymal stem cells on the 7 days.Result: The primary HUVECs and HDF had good morphology and strong proliferation, and the rBMSCs had a homogeneous long spindle shape, and the proliferation was strong. Under the microscope,HDF60%,HDF80% on the 7 days and HDF 40%,HDF 60% groups on the 14 days were arranged and woven into a dense network.CD31 immunofluorescence showed HDF60%,HDF80% was the most typical vascular network on the 7,14 days . Immunohistochemical staining showed that, on the 7 days, HDF 60% a,HDF 80% were closest to the content of type I collagen of human dermis . On the 1 days and 7 days, HDF 60% HDF 80% were closest to the content of type III collagen of human dermis. The flow cytometry showed that there were stem cells in every cell sheets of each group on the 7 days. Conclusion:rBMSCs,HUVECs,HDFs co -cultured with 9:5:21 (fibroblast 60% group) were cultured on the 7 days could formed cell sheets that had some properties.
