HUVEC细胞氚水长期暴露后的毒性效应

摘要:目的:探讨氚水染毒后人脐静脉血管内皮细胞(HUVEC)形态、大小、增殖、衰老改变。方法:HUVEC细胞用氚水连续染毒15代,普通光学显微镜下观察染毒细胞形态,Image J软件测量细胞表面积大小;CCK-8试剂盒检测不同时点HUVEC细胞增殖活性改变,β-半乳糖苷酶染色试剂盒检测氚水染毒后HUVEC细胞衰老的情况;COOLER算法计算氚水吸收剂量。结果:氚水染毒后,随染毒时间增加,HUVEC细胞形态发生改变,胞浆颗粒物增加,边界模糊;细胞表面积增加;T3、T6代细胞中观察到细胞衰老现象;T6、T9、T12、T15代细胞的增殖活性受到抑制;随着染毒时间增加,氚水所致细胞吸收剂量增加。可见,氚水对HUVEC细胞具有细胞毒性。

Toxic effects on HUVEC cells after exposure to HTO
Abstract:Objective: To investigate the changes of morphology, size, proliferation and senescence of HUVEC cells after exposure to tritium water. Method: HUVEC cells were exposed to tritium water for 15 consecutive generations. The morphology of cells exposed to tritium water was observed under ordinary optical microscope, the surface area of the cells was measured by Image J software. CCK-8 kit was used to detect the change of HUVEC cell proliferation activity at different time points, and β -galactosidase staining kit was used to detect HUVEC cell senescence after tritium water exposure. COOLER algorithm was used to calculate the absorbed dose of tritium water. Results: After exposure to tritium water, the morphology of HUVEC cells changed with the time of exposure, the cytoplasmic particles increased and the boundary was blurred. Cell surface area increased.Cell senescence was observed in T3 and T6 cells.The proliferation activity of T6, T9, T12, T15 cells was inhibited. With the increase of exposure time, the absorbed dose of cell induced by HTO increased. It can be seen that tritium water has cytotoxicity to HUVEC cells.
