血红素加氧酶-1的非酶活性研究进展

摘要:血红素加氧酶-1(HO-1)是体内参与血红素代谢的一种具有多种功能的微粒体氧化酶,具有极强防御氧化应激作用。然而近年来发现,HO-1具有酶活性以外的功能。HO-1可以从内质网转位至细胞核,参与调控抗氧化基因、炎症基因、干扰素诱导相关基因和癌基因等的转录,还可参与DNA修复和自身反馈调节,对多种疾病进程发挥重要的影响。本文将对HO-1非酶活性的作用研究现状进行综述。

The non-enzymatic activity of heme oxygenase-1
Abstract:IHeme oxygenase-1 (HO-1) is a microsomal oxygenase responsible for the catabolism of heme from destabilized heme proteins. For its potent anti-oxidant properties, HO-1 is regarded as a key cellular 20 defense mechanism. Recently, the effects of HO-1 independent of its enzyme activity have been identified. HO-1 is able to be transported from endoplasmic reticulum to the nucleus. After nuclear transportation, HO-1 regulates transcription of target genes responsible for redox oxidation, inflammation, interferon function and oncogenesis, and participates in DNA repairment and self-regulation. Thus, the enzyme activity-independent effect of HO-1 might play a critical role in the 25 pathogenesis of several diseases. The present article will review the current studies regarding the enzyme activity-independent effect of HO-1.
