细胞3D培养在组织工程研究中的应用

摘要:近年来,三维(3D)培养技术逐渐成为细胞生物学与组织工程学等交叉领域研究的热点之一,其要点是利用生物相容性高、良好生物降解性的材料,制备出所需的多孔架构,在体外构建出类似体内细胞生长依托的空间及基质环境,从而使得细胞呈现三维立体的生长,并表现出不同于平面二维培养的特征。细胞的3D培养为深入研究细胞生长及行为特性,尤其是肿瘤细胞的行为,以及干细胞增殖与分化这个组织修复的关键因素提供了良好的研究平台,将有望成为在基础医学体外研究过程中,传统的平面二维培养与临床体内研究的一个过渡与补充。本文主要从三维培养的特点、三维培养在组织工程(包括肿瘤研究)以及组织修复方面的研究与应用进行了阐述。

The application of cell 3D culture in the study of tissue engineering
Abstract:In recent years, the three-dimensional (3D) culture of cells has gradually become one of the hotspots in the cross field of cell biology and tissue engineering. The main point is to make use of the material with high biocompatibility and good biodegradability to prepare the required porous structure, and to construct a space like cell growth dependent in vitro. The substrate environment enables cells to grow in a three-dimensional way and shows characteristics different from two-dimensional planar culture. 3D cell culture provides a good research platform for the in-depth study of cell growth and behavior characteristics, especially the behavior of tumor cells, as well as the key factors for stem cell proliferation and differentiation, and will be expected to become a transition between traditional planar two-dimensional culture and clinical research in the process of basic medicine in vitro. This article reviews the characteristics of three-dimensional cell culture, and its applications in tissue engineering, tumor research and regenerative medicine.
