酿酒酵母SPT8基因的过表达载体构建

摘要:酿酒酵母发酵生产乙醇过程中,乙醇含量的不断增加,影响了酵母细胞膜的流动性并对膜蛋白产生毒性,这会使细胞的生长受到抑制甚至死亡,不利于乙醇的生产。因此,为研究酿酒酵母SPT8基因是否影响酿酒酵母细胞的乙醇耐性,本论文将酿酒酵母穿梭质粒pUG6上依次连入强启动子PGK、终止子CYC1以及目的基因SPT8,构建过表达载体pUPST。本论文为下一步研究SPT8基因是如何影酿酒酵母乙醇耐性的研究奠定了基础。也为研究工业酿酒酵母的乙醇耐性、提高发酵燃料乙醇的经济和技术水平奠定了基础。

The construction of overexpression vector of saccharom cerevisiae\'s SPT8 gene
Abstract:In Saccharomyces cerevisiae,high ethanol content influences the membrane fluidity and has toxic response for membrane protein during producing ethanol.This phenomenon results in cellar\'s growth inhibition or even death,which is not beneficial to the production of ethanol.Therefore,in this study,we use yeast\'s shuttle plasmid pUG6 to construct the overexpression plasmid pUPST by connecting the strong promoter PGK,terminator CYC1 and target gene SPT8 to study yeast\'s gene SPT8 whether effects yeast ethanol tolerance.This study lays the foundation that gene SPT8 is how to influence yeast ethanol tolerance,and of studying Industrial yeast ethanol tolerance,and of improving the level of economic and technical fuel ethanol fermentation.
