不同剂量荞麦对高胆固醇症小鼠的影响

摘要:目的:研究用不同剂量的荞麦粉代替高荞麦对高胆固醇症小鼠的影响胆固醇饲料中的麦芽糖糊精和玉米淀粉后对小鼠血液的脂质和胆固醇代谢相关基因的变化。方法:以AIN-93M标准饲料,高胆固醇模型饲料,高胆固醇和低剂量荞麦饲料以及高胆固醇和高剂量荞麦饲料等四种饲料作为C57BL / 6小鼠的饲料,通过对小鼠血清生化指标(总胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白)、肝脏组织形态学观察,肝脏胆固醇代谢基因HMG-CoA、srebp-2、CYP7A1、LXR-ɑ和 LDL-r等五个基因的相对表达量来分析荞麦的降胆固醇作用。结果:与模型组相比,荞麦组高低剂量组均能显著性降低血清总胆固醇,甘油三酯和低密度脂蛋白。与模型组和低剂量组相比,荞麦高剂量组显著降低了肝脏匀浆中的低密度脂蛋白。荞麦两剂量组均下调了HMG-CoA的相对表达量,上调srebp-2的相对表达量。与低剂量组相比,高剂量荞麦饮食还降低了LXR-ɑ的相对表达量,并且升高了LDL-r的相对表达量。结论:这些数据表明,荞麦的消费可能有助于预防和治疗高胆固醇血症。

Effects of different doses of buckwheat on hypercholesterolemia mice
Abstract:[Objective] Investigate the changes in the lipid and cholesterol metabolism related genes of C57BL/6 mice after the maltodextrin and corn starch replaced with different doses of buckwheat flour in high-cholesterol diet. [Methods] The C57BL/6 mice were fed the AIN-93M standard diet, high-cholesterol model (Paigen) diet, high-cholesterol and low-dose buckwheat diet and high-cholesterol and high-dose buckwheat diet,then measured the serum biochemical index in mice (total cholesterol, triglycerides, high-density lipoprotein cholesterol (hdl-c), low density lipoprotein) and liver tissue morphology observation, the relative expression of liver cholesterol metabolism genes (HMG CoA, srebp-2, CYP7A1, LXR-a and LDL-r) to analyze buckwheat cholesterol-lowering effect. [Results] The high doses buckwheat group die effects of buckwheat on hypercholesterolemia mice of Buckwheat diet on Hypercholesterolemic Micet significantly lowered the LDL-C in the liver homogenate compared with model group and low doses group. The relative expression of HMG-CoA and srebp-2 liver gene was down-regulated and up-regulated in the low-dose group of buckwheat. Compared with the low dose group, high doses buckwheat diet downgraded the relative expression of LXR-a and raise LDL-r in addition. [Conclusion] These data suggests that the consumption of buckwheat may help prevent and treat hypercholesterolemia.
