制备亲水色谱提取莲子免疫调节活性低聚糖的方法研究

摘要:制备色谱技术作为天然产物分离纯化的有效手段,快速简便、产品纯度高。其在药物研究尤其是活性成分的分离纯化中正发挥着越来越重要的作用。本研究通过使用中压制备亲水色谱(MPLC)搭配蒸发光散射检测器(ELSD)与紫外全波长扫描检测器(DAD)联用技术从莲子中分离获得五个高纯度的低聚糖,其纯度分别用高效阳离子交换色谱法(CationAcid Cation Exchange)测定均可达到90%以上。莲子低聚糖均能增加小鼠脾淋巴细胞的数量,具有一定免疫调节功能。

Study on extraction of immunoregulatory oligosaccharides by hydrophilic chromatography
Abstract:Preparation chromatographic technology as an effective means of separation and purification of natural products, fast and simple, high product purity. It is playing an increasingly important role in drug research, especially in the separation and purification of active ingredients. In this study, five high-purity oligosaccharides were isolated from lotus seeds by using medium pressure preparative hydrophilic chromatography (MPLC) combined with evaporative light scattering detector (ELSD) and ultraviolet full-wavelength scanning detector (DAD). The purity can be more than 90% by high-performance cation exchange chromatography (Cation Acid Cation Exchange). Lotus seed oligosaccharides can increase the number of mouse spleen lymphocytes and have certain immune regulation functions.
