鞣花酸尿石素对高脂膳食诱导肥胖小鼠血脂及肠道菌群的影响

摘要:目的:比较鞣花酸和尿石素A影响高脂膳食诱导肥胖小鼠血脂及肠道菌群的差异。方法:雄性C57BL/6J小鼠用普通饲料(NC)和高脂饲料(HFD)饲喂9周,选取HFD组肥胖小鼠,分别用低剂量(10mg/kg/d)、高剂量(30mg/kg/d)鞣花酸和尿石素(20mg/kg/d)灌胃处理7周,测体重、体脂、肝重,代谢活动,血脂及血清抗氧化指标,并用16S rRNA基因扩增子高通量测序分析结肠内容物菌群构成。结果:鞣花酸高、低剂量组及尿石素组均能显著降低高脂饮食诱导小鼠的体脂率(P<0.05),增加其夜间活动量(P<0.05),提高血清高密度脂蛋白降低血清丙二醛(p<0.01),提高血清SOD(p<0.05),但低剂量鞣花酸作用更明显。在属水平上,肥胖小鼠结肠菌群Desulfovibrio显著性升高(P<0.01),而各处理组均可降低其含量,尿石素组具有显著性差异(P<0.05);各处理组可不同程度地增加Odoribacter、coprostanoligenes、xylanophilum、Ruminococcaceae_UCG-010和Subdoligranulum含量。结论:鞣花酸高低剂量和尿石素A均可改善高脂饮食诱导肥胖小鼠的血脂水平并调节其肠道菌群,但鞣花酸低剂量组效果最好。

Effects of ellagic acid,urolithin on serum lipids and intestinal flora in the fat mice induced by high-fat diet
Abstract:Objective: To compare the effects of ellagic acid, urolithin A on serum lipids and intestinal flora in fat mice induced by high-fat diet. Methods: Male C57BL/6J mice were fed normal diet (NC) and high-fat diet (HFD) for 9 weeks, then fat mice in the HFD group were selected and treated with low-dose (10mg/kg/d) and high-dose (30mg/kg/d) ellagic acid and urolithin A (20mg/kg/d) respectively for 7 weeks. Body weight, body fat, liver weight, metabolic activity, serum lipid and antioxidant indexes were measured, and 16S rRNA gene amplifiers were used for high-throughput sequencing to analyze the microbiota of colonic contents. Results: Both the high and low dose group of ellagic acid and the urolithin A group significantly decreased body fat rate (P<0.05), increased their nocturnal activity (P<0.05), increased serum high-density lipoprotein cholesterol, decreased serum malondialdehyde (P<0.01), and increased serum SOD (P<0.05), but the effect of HFDEL was more obvious. Conclusions: Both high and low dose of ellagic acid and urolithin A could improve the serum lipids and regulate the intestinal flora of fat mice induced by high-fat diet, but the effect was best in the low-dose group of ellagic acid.
