CGRP在神经病理性疼痛作用机制研究进展

摘要:CGRP作为一种内源性神经肽,参与了机体许多生理和病理过程。近年来研究发现CGRP在神经病理性疼痛的发展和维持过程中发挥了重要作用,神经损伤后,伤害性信息从外周传递至脊髓背角,从而诱导脊髓背角中CGRP的释放,激活CGRP受体,增加脊髓背角细胞兴奋性。激活的CGRP受体与Gαs以及 Gαq或β-抑制蛋白偶联,从而激活PKB/AKt信号通路及MAPK级联反应,表明CGRP参与神经病理性痛调制过程。抑制CGRP的生成或阻止其发挥作用,从而抑制痛觉信息的信号转导进而缓解神经病理性疼痛。由此可探讨CGRP在神经病理性疼痛发生机制中的作用和地位。

Modulation Effect of CGRP in Neuropathic Pain
Abstract:IAs an endogenous neuropeptide, CGRP is involved in many physiological and pathological processes in the body. Recent studies have found that CGRP plays an important role in the development and maintenance of neuropathic pain. Nociceptive information is transmitted from the periphery to the spinal dorsal horn after nerve injury. Thereby inducing the release of CGRP in the spinal dorsal horn and activating CGRP receptors increase the excitability of spinal dorsal horn cells. Activation of the CGRP receptor coupled with Gαs and Gαq or β-arrestin activates the PKB/AKt signaling pathway and the MAPK cascade reaction, indicating that CGRP participates in the neuropathic pain modulation process. Inhibiting the production of CGRP or preventing it from functioning thereby inhibiting signal transduction of pain information and thus alleviating neuropathic pain. Therefore, the role and status of CGRP in the pathogenesis of neuropathic pain can be explored.
