N-乙酰半胱氨酸(NAC)对Ⅰ型糖尿病模型小鼠肝脏损伤的影响研究

摘要:目的:探究在1型糖尿病小鼠建模前进行N-乙酰半胱氨酸处理,对其成模后机体的氧化应激及肝脏损伤是否有缓解作用,从而为延长糖尿病模型动物的寿命提供指导意义。方法:将小鼠分为正常对照组(NC组)、正常对照+NAC处理组(NC+NAC组)、糖尿病组(DM组)、糖尿病+NAC处理组(DM+NAC组:在建立糖尿病模型前一周开始NAC干预)。每周检测动物的体重、血糖水平;八周后处死小鼠,收集血浆和肝脏组织,制作组织病理切片,测量肝脏氧化损伤指标ROS和MDA,及血清中ALT、AST和LDL指标。 结果:跟DM组相比,DM+NAC组小鼠体重偏重、而血糖偏低。比较肝脏组织MDA、ROS水平发现:DM+NAC处理组较未处理组有显著性差异(p<0.05);肝脏组织病理切片表明:DM小鼠肝脏组织出现肝索紊乱、肝细胞呈泡样病变,而DM+NAC组则为肝索以肝窦为中心呈放射状排列的现象。血清AST、ALT及LDL结果显示:AST、ALT在DM+NAC处理组中的含量显著低于DM组,与其他组没有显著差异。结论:在建模前用NAC处理的小鼠可以降低成模后其体内的氧化应激,缓解炎症,且血糖水平相对较低,说明肝脏生理状态影响1型糖尿病小鼠模型的构建。

Effect of N-Acetylcysteine (NAC) on hepatic injury in type I diabetes mice model
Abstract:Objective: To explore the effect of N-acetylcysteine treatment before modeling in type 1 diabetic mice, whether it alleviates the mice\'s oxidative stress and liver injury after modelling, thus provide guidance significant for prolonging the life span of diabetic mice model. Methods: The mice were divided into normal control group (NC group), normal control group+NAC (NC+NAC group), diabetes group (DM group), diabetic+NAC group (DM+NAC group: NAC intervention begins one week before diabetic model was established). Body weight and blood glucose level of mice were measured every week. The mice were sacrificed after NAC continuous intervention for eight weeks. Plasma was collected to detect the contend of AST、ALT and LDL, Liver tissues were used to made histopathological sections, and the contend of ROS and MDA in hepatocytes were measured with kit. Results: Compared with the DM group, before establishing diabetic mice model with NAC treated had a trend of weight gain and lower blood sugar. Comparison of MDA and ROS levels in hepatocyte showed a significant difference between the diabetic mice group and before establishing diabetic mice model with NAC treated group (p<0.05). Histopathological examination of the liver showed that hepatic cord disorder and hepatocyte appearance were observed in liver tissue of DM mice. And before the DM establising with NAC treated group, the hepatic cord was arranged radially with the liver sinus as the center. The results of serum AST, ALT and LDL showed that the content of AST and ALT in the group with NAC treated before DM mice model establishing was significantly lower than DM group, which was not significantly different from other groups. Conclusions: With NAC-treated before mice modeling can reduced oxidative stress, inflammation, and relatively low blood glucose levels , suggesting that liver physiological conditions influence the construction of type 1 diabetic mice model.
