小菜蛾p53基因的克隆表达及其功能的初步研究

摘要:p53基因是维持机体基因组稳态的重要基因。本项目以小菜蛾为研究材料,以期通过分子生物学等方法,研究细胞应激因子P53的功能。小菜蛾p53基因的全长2647bp,Genbank登录号为KF991220 ,其中编码区长1128bp,编码375个氨基酸。在此基础上,对其进行了生物信息学的表征,如保守结构分析,同源性分析等。本研究对小菜蛾p53基因进行了原核表达,蛋白约40-44KD,与预测大小相符。另外,利用昆虫表达载体在小菜蛾离体细胞中能成功表达小菜蛾p53基因。过表达P53基因对下游凋亡基因reaper和gadd45有一定的诱导作用,但是并不能引起明显的细胞凋亡。本研究通过不同的凋亡诱导因素对小菜蛾离体细胞进行凋亡诱导,发现p53基因在早期(2h)有一定的高表达,但在中后期(6h-12h)表达量迅速下降。同时发现小菜蛾p53基因主要在蛹期和成虫期高表达,在卵期和老熟幼虫期表达量较低。而其负调控因子MDM2主要在卵期高表达,在成虫期低表达,在蛹期几乎不表达。通过计算机模型也显示P53蛋白的C-端能与MDM2的N-端结合,这也间接反映了其互作关系。总之,本研究在成功克隆并表达小菜蛾p53基因的基础上,对其功能进行了初步的功能研究,揭示其可能存在的调控网络。

The cloning, expression and preliminary function study of p53 in Plutella xylostella
Abstract:The p53 gene is the key gene to protect the stability of genome in multicellular organisms. The aim of present paper is to study the primary function of the p53 gene in diamondback moth with molecular biology methods, so as to reveal the control pathway of apoptosis in this model organism. The full-length of p53 is 2647 bp with a CDS of 1128 bp which is coding a protein of 375 aa. The Genbank accession number of Plutella xylostella p53 gene is KF991220. Based on this, the bioinformatic characterization such as conserved domain of protein and phylogenetic analysis, was performed. The prokaryotic expression of p53 showed that the P53 protein is about 40-44KD. And the eukaryotic expression of p53 gene in cultured cell line of Plutella xylostella showed that the overexpression of p53 can elevate the expression of its downstream target apoptotic genes such as reaper and gadd45, and can induce apoptosis slightly but not obviously. After the induction of apoptosis with botanical pesticides reagents, the expression of p53 is elevated to a certain extent in treatment of 2 hours, while reduced sharply after 6 hours and 12 hours of the treatments, when comparing with the control. The qRT-PCR results showed that p53 was expressed mainly in pupae and adult stage, merely in egg and last instar larvae. While the expression pattern of MDM2, the negative regulator of p53, seemed somehow opposite, expressing highly in egg, but lowly in adult and merely in pupae. Just like reported previously in numerous literatures, our computer model also showed that the P53 binding the MDM2 with the C-terminus. Totally, our project study the primary function of p53 gene in Plutella xylostella, based on the cloning and the relationship between p53 and its putative target genes.
