A validation chiral HPLC method for the enantiomeric separation of Melphalan HCl

Abstract:A new and accurate chiral liquid chromatographic method was described for the enantiomeric separation of Melphalan HCl[L-3-[p-[Bis(2-chloroethyl)amino]phenyl]alanine hydrochloride]. The enantiomers of Melphalan HCl were baseline resolved on a (S,S)-Whelk-01, 5/100 Kromasil (250mm×4.6 mm, 5μm) column using a mobile phase system containing Methanol: Water (45:55, v/v). The resolution between the enantiomers was not less than 2 and interestingly distomer was eluted prior to eutomer. The limit of detection and limit of quantification of D- 3-[p-[Bis(2-chloroethyl)amino]phenyl]alanine hydrochloride [(D)-isomer] were found to be 0.08 and0.25μg/mL, respectively, for 10μl injection volume. The percentage recovery of (D)-isomer ranged from 91.5 to 95.0 in the bulk drug sample. The validated method yielded good results regarding precision, linearity, accuracy and ruggedness. The proposed method was found to be suitable and accurate for the quantitative determination of (D)-isomer in bulk drug samples of Melphalan HCl.

摘要:新建立了一种准确高的手性高效液相色谱方法,分析美法仑盐酸盐[L-3-[对-[双(2-氯乙基)氨基]苯基]丙氨酸盐酸盐]的对映体。美法仑盐酸盐的对映体分离使用(S,S)-Whelk-01 (250mm×4.6mm,5μm)色谱柱,流动相为甲醇:水(45:55,v/v)。对映体间之间的分离度不小于1.5,而且对映体在对映体形成之前就被洗脱了。分别注入10μL体积,检测D-3-[对-[双(2-氯乙基)氨基]苯基]丙氨酸盐酸盐(D型异构体)的检测限和定量限分别是0.08 和0.25μg / ml。(D)-异构体在美法仑盐酸盐样品中加样回收率在91.5 ~ 95.0之间。验证结果表明,该方法在精度、线性度、精度和耐用性等方面均取得了较好的效果。结果表明,该方法适用于美法仑盐酸盐样品中(D)-异构体的定量测定。
