小鼠急性心肌梗死模型的建立及心功能评价

摘要:目的通过结扎左前降支冠状动脉成功建立小鼠心肌梗死模型,并使用心脏超声进行心功能评价分析。方法 C57BL/6雄性小鼠30只,进行小动物无创气体麻醉,采用左侧肋间手术路径进入左胸腔,结扎冠状动脉左前降支,建立急性心肌梗死模型。术后3周使用小动物心脏超声评价左室收缩末期直径、舒张末期直径,射血分数和缩短分数。术后3周超声检查完成后取心脏标本病理学检查。结果小鼠心梗模型建模术后3周内死亡率13.3% (4/30),使用超声检查评价,建模成功率86.7%(26/30)。建模后小鼠心功能明显下降,心室明显增大,病理切片可见大量心肌细胞坏死,瘢痕形成。结论采用结扎冠脉左前降支可以成功建立小鼠心肌梗死模型并可用无创超声评价心功能。

Establishment of animal model for acute myocardial infarction and assessment of cardiac function
Abstract:(Objecyive) Establish an animal model of myocardial infarction and assess cardiac function by echocardiography. (Methods) Thirty male C57BL/6 mice used in this study. The left anterior descending coronary artery was ligated after anesthesia and intubation under the microscope. The echocardiographic assessment of myocardial infarction and cardiac function was performed after operation 4 weeks. Left ventricular end systolic diameter (LVESD), left ventricular end diastolic diameter (LVEDD), ejection fraction (EF) and shartening fraction (FS) were measured and nanlyzed. (Results) The acute mortality was 13.3% (4/30) during the model establishment. The echocardiography showed that the success rate of establishment of the infarction model was 86.7% (26/30). Cardiac function marker EF/FS anEstablishment of animal model for acute myocardial infarction and assessment of cardiac functiond decreased significantly, LVESD and LVEDD were increased. Pathological examination confirmed the scar formation and the necrosis of cardiac myocytes. (Conclusion) Murine myocardial infarction model was established successfully by ligation of left anterior descending coronary artery.
