化痰活血方对哮喘小鼠ILC2s相关因子的影响

摘要:目的以ILC2为核心研究化痰活血方(Huatan Huoxue Fang,HTHXF)对卵清蛋白(Ovalbumin,OVA)诱导的哮喘小鼠肺泡灌洗液(Bronchoalveolar lavage fluids,BALF)中IL-33、TSLP、IL-9、IL-6、AREG和血清中OVA-IgE表达的影响。方法雌性BALB/c小鼠40只随机分为正常组(C)、模型组(M)、地塞米松组(D)、化痰活血方低剂量组(HTHXF-L)和化痰活血方高剂量组(HTHXF-H)共5组,8只/组。于实验第1,8,15d除正常组腹腔注射生理盐水外其余各组腹腔注射OVA混悬液(含OVA1mg /ml,氢氧化铝5mg /ml)0.2ml致敏,第21-27d超声雾化吸入2%OVA激发液诱导哮喘小鼠模型。于实验第9-27d灌胃给药,雾化激发期间前1h灌胃给予各组药物。实验第28 d处死小鼠,分别获取BALF和血清,采用ELISA法测定哮喘小鼠BALF中IL-33、TSLP、IL-9、IL-6、AREG和血清中OVA-IgE(Immunoglobulin E)。结果化痰活血方能明显降低哮喘小鼠BALF中IL-33、TSLP、IL-9、IL-6和AREG的水平(P<0.01),并显著下调血清中OVA-IgE的水平(P<0.01)。结论化痰活血方治疗哮喘的作用机制可能与抑制ILC2s相关因子的分泌,继而调控Th2型免疫反应有关。

Effect of Huatan Huoxue Fang on ILC2s related factors in asthmatic mice
Abstract:Objective Using ILC2 as the core to study the effects of Huatan Huoxue Fang (HTHXF) on the levels of IL-33,TSLP,IL-9,IL-6,AREG in Bronchoalveolar lavage fluids (BALF) and OVA-IgE in serum of ovalbumin (OVA)-induced asthmatic mice.Methods 40 female BALB /c mice were randomly divided into 5 groups (8 mice / group), including normal group (C), model group (M), dexamethasone group (D), low dose group (HTHXF-L) of Huatan Huoxue Fang and high dose group (HTHXF-H).The asthmatic mice were sensitized by intraperitoneal injection of 0.2 ml of OVA suspension (containing OVA 1mg/ml, aluminum hydroxide 5mg/ml) on the 1st, 8th and 15th day except normal group. The asthmatic mice were induced by inhaling 2% OVA stimulant on the 21st to 27th day. The mice were given intragastric administration at 9-27d, and the mice were given intragastric administration before 1h of ultrasonic atomizer. On the 28th day of the experiment, BALF and serum were obtained. The levels of IL-33, TSLP, IL-9, IL-6, AREG in BALF and OVA- IgE (Immunoglobulin E) in serum of asthmatic mice were determined by ELISA. Results HTHXF could significantly reduce the levels of IL-33, TSLP, IL-9, IL-6 and AREG in BALF of asthmatic mice (P < 0.01), and significantly reduce the levels of OVA- IgE in serum (P < 0.01). Conclusion The mechanism of Huatan Huoxue Fang in the treatment of asthma may be related to the inhibition of the secretion of ILC2s related factors , and then the regulation of Th2- type immune response.
