预先低剂量微波辐射增强γ射线诱导小鼠白血病L1210细胞凋亡

摘要:目的:研究预先低剂量微波辐射对γ射线诱导L1210细胞凋亡的影响。方法:将小鼠白血病L1210细胞随机分为对照组(C)、单纯微波组(RF)、单纯γ射线组(γ)和联合暴露组(RF+γ)。单纯微波组和联合暴露组给予120μW/cm2微波照射,每天4h,连续辐照5d;第5d给予单纯γ射线组和复合组8Gy γ射线照射。用钙离子依赖性磷脂结合蛋白-异硫氰酸荧光素/碘化丙啶(Annexin V-FITC/PI)双标记方法检测细胞凋亡,通过Western-blot检测Survivin蛋白的表达水平。结果:与对照组相比,单纯γ射线组凋亡率明显升高(P<0.05);与单纯的γ射线组相比,联合暴露组凋亡率显著升高(P<0.05)。与对照组相比,单纯γ射线组细胞内Survivin蛋白表达水平明显降低(P<0.05)。与单纯γ射线组相比,联合暴露组Survivin蛋白表达水平明显降低(P<0.05)。结论:本实验条件下,8Gyγ射线可以引起小鼠白血病L1210细胞凋亡率增加,Survivin蛋白表达水平降低。预先900MHz,120μW/cm2微波辐射能够显著增强γ射线引起的小鼠白血病L1210细胞的损伤,表现为细胞凋亡率增加和Survivin蛋白表达水平降低。

Pre-exposure of L1210 cells to 900MHz microwave radiation enhances apoptosis induced by γ-rays radiation
Abstract:Object: In this paper, the role of low dose microwave radiation on γ-rays induced L1210 cell apoptosis and the underlying mechanism were investigated. Methods: L1210 cells were radomly divided into control group, microwave alone group,γ-rays alone group and combined exposure group. Microwave alone and combined exposure groups were exposed to 120μW/cm2 microwave, 4h/d, for 5 days. The combined exposure group and γ-rays alone group were subsequently exposed to 8Gy 60Co γ-rays on 5th day. Apoptosis was detected with Annexin V-FITC/PI double labelling method. Survivin gene expression was measured with western blot. Results: Compared with control group, apoptosis of cells in γ-rays alone group significantly increased(P<0.05). Apoptisis rate of cells in combined exposure group was statistically higher than that of γ-rays alone group(P<0.05). Compared with control group, surviving gene expression level of γ-rays alone group decreased significally(P<0.05). Survivn gene expression of combined exposure group was statistically higher than that of γ-rays alone group(P<0.05). Conclusion: Under this experimental condition, 8Gy γ-rays increased apoptosis and decreased Survivin gene expression in L1210 cells. Pre-exposure with 900MHz, 120μW/cm2 microwaves could significantly increased the γ-rays induced damages to L1210 cells, as evidenced by the increase of apoptosis and decrease of Survivin gene expression.
