酶法合成富含1,3-二油酸-2-棕榈酸甘油三酯及1-油酸-2-棕榈酸-3-亚油酸甘油三酯的油脂组合物

摘要:在无溶剂体系中,以棕榈硬脂和油酸及亚油酸为原料,酶法合成富含1,3-二油酸-2-棕榈酸甘油三酯(OPO)和1-油酸-2-棕榈酸-3-亚油酸甘油三酯(OPL)的油脂组合物。探究酶的种类,底物摩尔比,加酶量,温度和反应时间对OPO和OPL的含量,sn-2位棕榈酸的含量及sn-2位棕榈酸占总棕榈酸的百分比的影响。在最优条件下(底物摩尔比,棕榈硬脂/油酸/亚油酸=1:8:4;NS40086添加量,8%;反应温度,60℃;反应时间,4h),得到OPO和OPL的总含量为69.26%,sn-2位棕榈酸的含量为87.75%,sn-2位棕榈酸占总棕榈酸的百分比为71.07%。该油脂组合物模拟人乳中主要的甘油三酯组成,同时提供必需脂肪酸,可应用于婴儿配方食品中。

Enzymatic synthesis of structured lipids rich in 1,3-dioleoyl-2-palmitoylglycerol and 1-oleoyl-2-palmitoyl-3-linoleoylglycerol
Abstract:Structured lipids, rich in 1, 3-dioleoyl-2-palmitoylglycerol (OPO) and 1-oleoyl-2-palmitoyl-3-linoleoylglycerol (OPL), were prepared by enzymatic acidolysis. The effects of enzyme type, substrate molar ratio, enzyme loading, temperature, and reaction time on the yield of OPO and OPL, palmitic acid (PA) content at the sn-2 position of TAG, and the percentage of PA at the sn-2 position relative to total PA content were investigated. Under optimal reaction conditions (palm stearin/oleic acid/linoleic acid molar ratio of 1/8/4, NS40086 loading oEnzymatic synthesis of structured lipids rich in 1,3-dioleoyl-2-palmitoylglycerol and 1-oleoyl-2-palmitoyl-3-linoleoylglycerolf 8%, 60 C, 4 h), the combined yield of OPO and OPL was 69.26%, PA content at the sn-2 position was 87.75%, and the relative percentage of PA at the sn-2 position was 71.07%. This lipid composition is similar to that found in human milk and provides essential fatty acids, thus can be useful in infant formulas.
