膀胱癌尿液蛋白质组学质谱研究方法

摘要:膀胱癌的诊断方法主要为尿脱落细胞学及膀胱镜检查,但尿脱落细胞学检查敏感度较差,而膀胱镜为有创检查,常给患者带来很大痛苦,因此临床上亟需寻找具备易于检测、诊断准确率高、可动态监测特点的尿液肿瘤标志物。尿液蛋白质组学研究是寻找生物标志物的有效手段,但由于蛋白质组学的固有复杂性、研究方法多样和结果混杂的特征,人们对其认识仍很模糊,尚需借助最先进的质谱技术不断探索完善。本文针对质谱检测过程中方法较为灵活可变的步骤,即从尿液标本的收集与保存、总蛋白质提取及质谱数据采集三个方面就膀胱尿路上皮癌尿液蛋白质组研究的具体实验方法进行简要评述,以期为临床和科研工作者提供理论参考。

Urinary proteomics strategies for urothelial bladder cancer: in mass spectrometry-based
Abstract:Urothelial bladder cancer is one of the most common urinary tract carcinomas. The earlier this cancer is found and treated, the better the outcome. UBC detection and surveillance include cytology, which suffers from low sensitivity, particularly for low-grade tumors, and cystoscopy, which is invasive and expensive. There is therefore a clinical need for biomarkers via a quick urine test to rapidly and accurately diagnose UBC Review of urinary proteomics strategies for urothelial bladder cancer: in mass spectrometry-basedand its relapse. Urine proteomic research is an effective way to find candidate biomarkers, which has been explored for many years. Based on the inherent complexity of proteomics and diverse methods of mass spectrometry analysis, existing research results from different groups are often confused and not easily referenced. Hence, with the development of mass spectrometry technology, this field still need to be explored constantly in new candidate finding and verification. In this review, three steps of mass spectrometry analysis, collection and preservation of urine specimens, total protein extraction and mass spectrometry data collection, involved in urine proteome analyzing of urothelial bladder cancer were mentioned and discussed to provide a theoretical reference for clinical and scientific researchers.?????
