高通量测序研究西双版纳植物叶际真菌种群

摘要:为探明西双版纳植物叶际真菌多样性,应用高通量测序分析方法,对7个随机调查采样点39个植物样品进行检测,共获得9,178,355对 Reads,每个样品叶际真菌OTU数量在365-1878之间,采样点之间和样品间均存在共同的真菌种群,也有明显的差异。草本与木本植物都具有很高的叶际真菌多样性。主要检测到3个主要门、10个纲、8个主要目、35个种真菌及归类真菌。采样点检出的真菌属种存在明显差别,每个植物样品都具有其独特的叶际真菌种类。研究检测到Cladosporium_delicatulum、Hyphopichia_burtonii、Uwebraunia_dekkeri、Trimmatostroma_cordae为植物与地点分布普遍的真菌,Filobasidium_sp,Meira_nashicola,Pyrenochaetopsis_leptospora等局部植物与地点分布的真菌。叶际真菌中包括潜在的植物致病菌、生物防治菌,但多数叶际真菌的功能尚待研究。本研究为探索植物病原真菌的溯源、流行,叶际真菌对热带雨林生态稳定性的作用及开发应用有益真菌奠定了基础。

High-throughput sequencing study on Plant Phyllosphere Fungal Population in Xishuangbanna,China
Abstract:In order to explore the phyllosphere fungal population of Xishuangbanna, high-throughput microbial diversity analysis was applied to sequence 39 healthy plant samples without diseased symptoms at 7 random survey sampling plots. A total of 9,178,355 pairs of Reads were obtained and the number of fungal OTU was 365-1878 per sample. OTU analysis shows that there is a common fungal population among the phyllosphere fungi in Xishuangbanna, which differs not only from each sampling point, but also from plant samples. analysis also showed that both herbaceous and woody plants had high fungal diversity. Our study indicates that both woody and herbaceous plants in Xishuangbanna harbor diverse phyllosphere fungi. Three main phylums, ten classes, eight main orders, 35 species and fungi were detected. There are obvious differences in fungi species detected at sampling points and each plant sample has its own unique species of phyllosphere fungi. Cladosporium_delicatulum、Hyphopichia_burtonii、Uwebraunia_dekkeri、Trimmatostroma_cordae that commonly distributed in plants and places, and Filobasidium_sp, Meira_nashicola, Pyrenochaetopsis_leptospora that locally distributed in plants and places were detected. The function of detected fungi include potential plant pathogens and some biological control fungi, but the most of phyllosphere fungi whose functions are still to be explored. This study laid a foundation for exploring the origin and epidemic of plant pathogenic fungi, the role of phyllosphere fungi in the ecological stability of tropical rainforest and the development and application of beneficial fungi.
