IDH1突变对胶质瘤干细胞辐射敏感性的影响

摘要:目的:探讨异柠檬酸脱氢酶1(Isocitrate dehydrogenase1,IDH1)基因突变对胶质瘤干细胞(Glioma stem cells,GSCs)辐射敏感性的影响。方法:通过全自动流式细胞分选技术,依据GSCs表面干性标志物CD133,富集培养细胞。构建IDH1 突变过表达质粒转染GSCs,Western blot技术检测突变蛋白表达水平。通过CCK8实验检测IDH1突变对GSCs增殖活力的影响,细胞免疫荧光实验检测照射后不同时间点γH2AX foci的数量,Western blot技术检测照射前后TIGAR蛋白表达水平,氧化还原试剂盒检测NADP+/NADPH及GSSG/GSH表达水平。结果:CCK8显示IDH1突变对GSCs增殖活力的影响无显著性差异。细胞免疫荧光实验表明对照组与实验组γH2AX foci形成的数量一致,Western blot显示照射引起TIGAR蛋白水平增加,IDH1突变未造成TIGAR蛋白表达量上升,氧化还原实验表明实验组还原性物质有所增高,但无统计学差异。结论:IDH1基因突变未造成GSCs增殖活力,DNA损伤修复能力、TIGAR蛋白表达和还原性物质水平下降,联合X射线照射后,IDH1 R132H对GSCs的放射敏感性没有明显影响。

The effects of IDH1 mutation on radiosensitivity of GSCs
Abstract:Objective: To investigate the effects of Isocitrate dehydrogenase 1 (IDH1) gene mutation on radiosensitivity of glioma stem cells (GSCs). Methods: GSCs were enriched and cultured by automated flow cytometry according to CD133, a stem marker of GSCs. The plasmid in which IDH1 gene mutation was overexpressed was constructed and transfected into GSCs, and the expression levels of the mutant protein were detected by Western blot. The effects of IDH1 mutation on the proliferation of GSCs were detected by CCK8 assay. The number of γH2AX foci at different time points after irradiation was detected by cell immunofluorescence technique. The expression of TIGAR protein was detected by Western blot. The redox kit detected the ratios of NADP﹢/NADPH and GSSG/GSH in the control and the experimental groups, respectively. Results: IDH1 mutation on the proliferation of GSCs was not significant by CCK8. Cellular immunofluorescence experiments showed that the amount of γH2AX foci formed in the control group was consistent with that in the experimental group. Western blot results indicated that irradiation caused an increase in TIGAR protein levels, but the amount of protein increased was basically same, The results of redox experiments showed that the reducing substances in the experimental group increased slightly, compared to the control group, but there was no statistical difference. Conclusion: IDH1 gene mutation did not decrease the GSCs` proliferation, DNA damage repair ability, TIGAR protein expression and reducing substances, and the radiation sensitivity ofIDH1 mutant GSCs was not increased after irradiation.
