表层蛋白对乳杆菌耐受性和黏附功能的影响

摘要:【目的】筛选携带表层蛋白的乳杆菌,探究表层蛋白对于菌株自身胃肠耐受性和黏附相关性质的影响。【方法】使用5 mol/L LiCl提取表层蛋白并通过SDS-PAGE对提取物进行电泳鉴定。通过活菌计数比较去除表层蛋白前后温育在模拟消化液中的菌株存活率。通过乳杆菌表面性质测定及对HT-29细胞黏附实验探究表层蛋白对菌株表面性质和黏附功能的影响。【结果】筛选出四株携带表层蛋白的乳杆菌L. acidophilus CGMCC 1.1878、L. fermentum CCFM 421、L. crispatus JCM 2009、L. gohnsnil JCM 1101。相较完整菌株,去除表层蛋白后菌株在模拟胃肠液中存活率显著下降。失去表层蛋白的菌株其自聚性、共聚性、疏水性、表面带电性以及对HT-29细胞的黏附率均显著低于完整菌株(p<0.05)。【结论】表层蛋白对胃肠道环境中的乳杆菌具有保护作用,对乳杆菌的胃肠耐受性、表面性质以及黏附功能具有积极贡献,是保证菌株存活并定植于宿主细胞的重要原因。

Effects of surface layer proteins on strains' tolerance and adhension function
Abstract:[Objective] The Lactobacillus carrying the surface layer proteins(Slps) was screened to investigate the effect of the Slps on strains' gastrointestinal tolerance and adhesion-related properties. [Method] 5 mol/L LiCl and SDS-PAGE were used to extract and identify the Slps respectively. Viable counts were used to compare strains' survival rate before and after removal the Slps. The effects of Slps on the surface properties and adhesion function of strains were investigated by measuring the surface properties of Lactobacillus and adhering to HT-29 cells. [Result] Four Slps extracted by L. acidophilus CGMCC 1.1878、L. fermentum CCFM 421、L. crispatus JCM 2009、L. gohnsnil JCM 1101 were identified. Compared with the intact strains, the survival rate of the strains in the simulated gastrointestinal environment was significantly decreased after the Slps were removed. The auto-aggregation, co-aggregation, hydrophobicity, surface charges and adhesion ability to HT-29 cells of the strains that lost the Slps were significantly lower than that of intact strains (p<0.05). [Conclusion] Slps have protective effects on Lactobacillus in the gastrointestinal environment, and have positive contributions to strains' gastrointestinal tolerance, surface properties and adhesion function. Slp is an important reason for Lactobacillus to survive and colonize the host cell.
