姜黄素的药理作用与不良成药因素分析

摘要:姜黄素是从郁金、姜黄、莪术等姜黄属临床常用中药中提取出来的具有双阿魏酰甲烷结构特征的多元酚类化合物,大量的临床与实验研究发现,姜黄素类化合物具有良好的抗肿瘤,抗炎和抗氧化等多种药理活性,临床应用广泛。有研究显示,姜黄素存在着降解速率快、生物利用度低等缺点,因而成药性较差,限制了姜黄素的进一步开发和利用。本文综述了姜黄素的基本化学性质和药理活性,同时也对姜黄素的不良成药因素进行了分析,并对基于姜黄素类化合物结构母核的药物设计与开发的未来研究思路与方向做出了展望。

Pharmacological activities and poor druggability factors analyses of Curcumin
Abstract:Curcumin is a series of polyphenols extracted from the traditional Chinese medicine genus Curcuma. Large amount and long-term clinical and experimental researches revealled that herbal medicines, such as Curcuma wenyujin Y, H. Chenet C. Ling, Curcuma longa L. and Curcuma zedoaria (Christm.) Rosc. in genus Curcuma have various pharmacological acvities, such as anti-tumor, 25 anti-inflammaton and anti-oxidation, etc. and are widely applied clinically. However, experiments and research data indicated that the druggabilities of curcumin compounds were poor, and scientists ascribed the disadvantages to the rapid degradation rate and low bioavailability. These weaknesses limited the further development and utilization of curcumin. In this paper, the basic chemical properties and pharmacological effects and the factors of poor draggabilities of curcumin compounds were 30 summarized. Then, after combining the advantages and disadvantages of curcumin, the future research direction of the drug design and development based on curcumin compounds scaffold is prospected.
