多肽类轮烷的固相合成探索

摘要:轮烷是超分子化学中重要的机械互锁分子,轮烷在材料科学、纳米技术以及生物科学等领域的广泛应用已越来越受到科学家的关注。轮烷的合成多是在液相中合成的,利用这种方法合成轮烷步骤繁琐,耗时长且经多次的分离提纯导致产率低。目前在固相上合成轮烷的研究报道还不多,本课题采用在固相上合成轮烷,因反应在一简单反应器皿即固相反应管中便可进行,其操作简便,易于纯化。因此在固相上合成轮烷对于轮烷的合成来说无疑是一种新颖的合成方法。另一方面,本课题的轴分子为氨基酸形成的短肽,环状分子为酰胺类大环,二者均具有较好的生物相容性,因此合成的轮烷有望成为构筑生物材料的基本单元。

Exploration of solid phase synthesis of polypeptide rotaxane
Abstract:Rotaxane is an important mechanical interlocking molecule in supramolecular chemistry. The widespread application of rotaxane in materials science, nanotechnology and biological sciences has attracted more and more attention from scientists. The synthesis of rotaxane is mostly synthesized in the liquid phase. The step of synthesizing the rotaxane by this method is cumbersome, time consuming and subjected to multiple separations and purifications resulting in low yield. At present, there are few research reports on the synthesis of rotaxane on the solid phase. This topic uses the synthesis of rotaxane on the solid phase. The reaction can be carried out in a simple reaction vessel, ie, a solid phase reaction tube, which is easy to operate and easy to purify. Therefore, the synthesis of rotaxane on the solid phase is undoubtedly a novel synthesis method for the synthesis of rotaxane. On the other hand, the axis molecule of this subject is a short peptide formed by amino acids, and the cyclic molecule is an amide macrocycle, both of which have good biocompatibility, so the synthesized rotaxane is expected to be the basic unit for constructing biomaterials.
