过氧天然产物共有1,2-二氧环己烷核心骨架的合成研究

摘要:1,2-二氧环己烷是过氧天然产物中最具代表性的结构,同时也是最具广泛生物活性的一类分子。本研究工作发现通过强Lewis酸或Bronsted酸活化3-烷氧基-1,2-二氧环己烷前体,产生高活性的过氧碳正离子中间体与一系列硅基烯酮缩醛等亲核试剂发生加成反应,从而实现了结构多样的1,2-二氧环己烷核心骨架的高效合成。本研究工作为后续plakortide家族及相似结构的过氧天然产物分子的通用合成奠定了良好的方法学基础。

Synthetic study towards 1,2-dioxane structure core of peroxy natural products
Abstract: 1,2-dioxanes are common structural motifs widely founded in numerous biologically active peroxy natural products. In this work, we successfully developed a novel strategy featuring the use of strong Lewis acid or Bronsted acid-promoted peroxidation reactions of highly reactive peroxycarbenium ion intermediates with series of silyl ketene nucleophiles, for the rapid access to a variety of important 1,2-dioxane scaffolds from the readily available 3-alkoxy-1,2-dioxane precursors. The new reactions established herein will provide an efficient method for the collective syntheses of plakortides and relating peroxy natural products.
