基于ITS序列对祁连山东段大型真菌的分子鉴定

摘要:我国拥有丰富的大型真菌资源,但是目前仍存在利用少、利用效率低的问题。本研究对祁连山东段青海云杉林内的15株大型真菌进行分子鉴定,通过提取子实体DNA,利用PCR技术对其rDNA的ITS区段进行扩增测序,与GenBank数据库中的序列进行BLAST比对,并对其进行系统发育分析。结果显示:它们全部属于担子菌门,13株属于伞菌目,1株属于非褶菌目,1株属于柄灰包目。本研究对当地大型真菌的多样性进行一定的探明,为野生大型真菌资源的利用和保护提供了参考依据。

Molecular Identification of macro-fungi in eastern Qilian Mountain
Abstract:China is rich in macro fungi resources, but there are still some problems such as low utilization efficiency.In order to make a preliminary study on the resources of macrofungi in China. In this study, 15 strains of macro-fungi in Picea crassifolia forest in the eastern Qilian Mountain were identified by molecular identification. The rDNA ITS region being extracted from the fruiting body, was amplified by PCR technique, sequenced and compared with the sequence in the GenBank database. The phylogenetic analysis results showed that all of them belonged to Basidiomycetes, 13 belonged to Agaricales, 1 to Polyporales, and 1 to Tulostomatales. The diversity of local macro-fungi was preliminary studied in this paper.This study provides a reference for utilization and protection of wild macro-fungi resources.
