Vero细胞微载体悬浮培养生产猪伪狂犬病毒工艺研究

摘要:微载体因具高比面积、易操作等优点被广泛应用于动物疫苗行业,国内市场需求量巨大。目前商业化微载体主要为进口微载体,价格昂贵,因而急需自主研发出国产微载体。猪伪狂犬病毒严重危害猪群的健康,给养殖户造成巨大的经济损失,目前疫苗免疫预防是控制该病爆发的主要有效方法。本研究成功的应用自制微载体悬浮培养Vero细胞,建立了一种安全可靠的猪伪狂犬病毒生产工艺。首先在125 mL磁力搅拌瓶中对微载体的浓度和细胞接种密度进行优化,确定Vero细胞的最适培养条件,然后进一步对PRV病毒培养工艺参数(接毒时间(TOI)、感染复数(MOI)、收毒时间)进行优化,将优化后的工艺参数在3 L反应器上进行规模放大验证。当以TOI为72 h,MOI为0.1,接毒72 h后,病毒滴度为7.3 LgTCID50omL-1,相比已报道文献中病毒的最大滴度提高了约6.3倍。本研究开发出的经济实用型微载体将会为工业生产动物疫苗以及PRV的规模化生产奠定基础。

Production of Pseudorabies Virus by Vero cells grown on dextran microcarriers
Abstract:Microcarriers have been widely applied in the animal vaccine industry because of its advantages, such as high specific surface area and easy to operate, and the market demand is large. At present, commercial microcarriers are mainly imported microcarriers. Due to their high price, it is urgent to independently develop domestic microcarriers. Porcine pseudorabies virus seriously harms the swinery health, causing huge economic losses to farmers. Vaccine immunization prevention is currently the main effective method to control the outbreak of the disease. In this study, self-made microcarriers suspension culture Vero cells were used to produce Porcine Pseudorabies virus, and a safe and reliable virus production process was established. Firstly, the concentration of microcarriers and Vero cell seeding density was optimized in a 125 mL magnetic stirring bottle to create optimal conditions for the self-made microcarriers to cultivate the Vero cells. Secondly, the process parameters of PRV virus culture were optimized, including the time of infection(TOI), multiplicity of infection (MOI), and harvest time. Thirdly, the optimized process parameters were scaled up in a 3 Liter Bioreactor. Based on 72 h cultivation of cells, the highest virus titer LgTCID50omL-1 value of culturing the PRV virus was 7.3 after being infected 72 h at MOI=0.01, which was 6.3 times higher than the reported literature. The economical and practical microcarriers developed in this study will lay the foundation for industrial production of animal vaccines and large-scale production of PRV.
