PER1对人肝癌细胞BEL-7402辐射敏感性的影响

摘要:目的:研究Per1在人肝癌细胞辐射敏感性中的作用。方法:X射线照射SMMC-7721、BEL-7402和HepG2肝癌细胞后,克隆形成实验检测其存活分数;Western blot和 RT-qPCR 检测Per1在肝癌细胞系中的表达;X射线照射过表达Per1的肝癌细胞,克隆形成实验检测存活分数;流式细胞仪检测凋亡。结果:在梯度剂量X射线0、2、4、6、8、10Gy照射下SMMC-7721,BEL-7402和HepG2克隆存活分数逐渐下降,其中SMMC-7721对辐射最敏感,BEL-7402辐射抗性最高。Western blot检测发现Per1在SMMC-7721中的表达水平明显高于BEL-7402和HepG2(p<0.05)。过表达Per1蛋白以后,BEL-7402接受5Gy X射线照射后凋亡明显增多,同时,Western blot 和RT-PCR实验结果发现,X射线照射过表达Per1的BEL-7402细胞,抗凋亡蛋白Bcl-2表达明显降低,促凋亡蛋白Caspase3表达明显升高。结论:PER1蛋白的高水平表达可以促进X射线诱导的凋亡,增强肝癌细胞的辐射敏感性。

Effect of PER1 on Radiation Sensitivity of Human Hepatoma Cell Line BEL-7402
Abstract:objective: To study the role of Per1 in the radiation sensitivity of human hepatocellular carcinoma. Methods: MMC-7721, BEL-7402 and HepG2 hepatoma cells were irradiated by X-rays, and the survival fraction was detected by colony formation assay. Western blot and RT-qPCR were used to detect the expression of Per1 in hepatocellular carcinoma cell lines and the expression of apoptosis-related proteins. Hepatoma cells overexpressing Per1 were irradiated by X-ray, and the survival fraction was detected by colony formation assay. Apoptosis was detected by flow cytometry. Results: The survival fraction of SMMC-7721, BEL-7402 and HepG2 clones decreased gradually under gradient dose X-rays of 0, 2, 4, 6, 8, 10 Gy. SMMC-7721 was the most sensitive to radiation and BEL-7402 radiation resistance highest. Western blot analysis showed that the expression level of Per1 in SMMC-7721 was significantly higher than that of BEL-7402 and HepG2 (p<0.05). After overexpression of Per1 protein, apoptosis of BEL-7402 was significantly increased after 5Gy X-ray irradiation. At the same time, Western blot showed that the expression of anti-apoptotic protein Bcl-2 was significantly decreased and the expression of pro-apoptotic protein Caspase3 was significantly increased by X-ray irradiation of Per1 overexpressing BEL-7402 cells. Conclusion: High expression of PER1 protein can promote apoptosis induced by X-ray and enhance the radiation sensitivity of liver cancer cells.
