摘要:目的:探究自噬引起的造血系统异常对消化系统的影响。方法:用PCR和凝胶电泳技术鉴定Atg7f/f; Vav-iCre小鼠基因型。获得小鼠的肠组织,测量其长度。利用HE染色技术检测小鼠胃、肠的组织形态结构。抽提小鼠胃、肠组织的RNA,并利用实时荧光定量(Q-PCR)技术检测端粒酶相关基因的的表达水平。利用Western blotting技术检测小鼠胃、肠组织中DNA损伤情况以及P15蛋白质的表达。结果:Atg7f/f;Vav-iCre模型小鼠构建成功。造血系统Atg7基因缺失后,肠组织的长度无明显变化。造血系统Atg7基因缺失后,HE染色显示,小肠绒毛数量未减少、结构无明显改变,胃组织结构未见明显病变。造血系统Atg7基因缺失后,小鼠胃、肠组织的端粒酶相关基因转录水平无明显变化。造血系统Atg7基因缺失后,小鼠胃、肠细胞的DNA损伤标志物γ-H2AX蛋白的磷酸化以及P15蛋白表达量无明显改变。结论:自噬引起的造血系统异常对消化系统无明显影响。

The effect of abnormal hematopoietic system induced by autophagy on the digestive system
Abstract:Objective:To investigate the effects of abnormalities of the hematopoietic system caused by autophagy on the digestive system. Methods: 1) PCR and gel electrophoresis were used to identify Atg7f/f; Vav-iCre mouse genotype. 2) Mouse intestinal tissue was obtained and its length was measured. 3) HE staining technique was used to detect the histomorphological structure of stomach and intestine of mice. 4) RNA was extracted from the stomach and intestine of mice, and the activity of telomerase was detected by real-time fluorescence quantitative (Q-PCR) technique. 5) The DNA damage and P15 protein expression in the stomach and intestine tissues of mice were detected by Western blotting. Results: 1) Atg7f/f; Vav-iCre model mice were successfully constructed. 2) After the loss of Atg7 gene in hematopoietic system, the length of intestinal tissue of KO mice did not change significantly. 3) After the loss of Atg7 gene in hematopoietic system, HE staining showed that the number of small intestine villi was not reduced and the structure was not significant changes, no obvious lesions in the stomach tissue structure. 4) After the loss of the Atg7 gene in the hematopoietic system, the telomerase-related gene transcription levels in the stomach and intestine tissues of the mice did not change significantly. 5) After Atg7 gene deletion in hematopoietic system, there was no significant change in the phosphorylation of the DNA damage marker γ-H2AX protein and the expression of P15 protein in mouse stomach and intestinal cells. Conclusion: Hematopoietic abnormalities caused by autophagy have no significant effect on the digestive system.
