GAS2/Calpain2轴通过IDH2调控CML细胞生长

摘要:目的:探究靶向生长抑制特异蛋白(GAS2)抑制慢性髓细胞白血病(CML)细胞生长的分子机制。方法:利用同位素标记相对和绝对定量(iTRAQ)技术比较过表达GAS2显性负性突变体(GAS2DN)的MEG-01细胞和对照细胞中的蛋白表达变化;过表达GAS2DN或GAS2后,使用Q-RT-PCR和western blot检测MEG-01、BaF3/BCR-ABL和K562细胞中异柠檬酸脱氢酶(IDH2)的表达状况;Q-RT-PCR比较健康供体和CML患者CD34+细胞中IDH2的表达;研究IDH2沉默对MEG-01细胞生长和集落生成能力的影响。结果:iTRAQ分析提示GAS2DN抑制IDH2的表达;在MEG-01和BaF3/BCR-ABL细胞中过表达GAS2DN可抑制IDH2的蛋白表达,在BaF3/BCR-ABL细胞中过表达GAS2则促进IDH2的表达,但IDH2的mRNA表达均不受影响;沉默Calpain2可部分"挽救"GAS2DN在K562细胞中导致的IDH2表达抑制;IDH2在CML患者CD34+细胞中表达较对照细胞显著增高,沉默IDH2显著抑制MEG-01细胞的生长和集落形成能力。结论:靶向GAS2可部分通过抑制IDH2表达而影响CML细胞的生长。?????

GAS2/Calpain2 axis modulates the growth of CML cells via IDH2
Abstract:Objective: To delineate the molecular mechanism of how targeting of growth arrest-specific 2 (GAS2) inhibits the growth of chronic myeloid leukemia (CML) cells. Methods: Isobaric tag for relative absolute quantitation (iTRAQ) was utilized to analyze the protein expression of MEG-01 cells overexpressing the dominant negative form of GAS2 (GAS2DN) compared to the control cells; isocitrate dehydrogenase, mitochondrial (IDH2) expression was measured by Q-RT-PCR and western blot in MEG-01 and BaF3/BCR-ABL cells with GAS2 or GAS2DN overexpression; K562/GAS2DN cells were transducted with lentiviral vector delivering shRNA against Calpain2, and then IDH2 expression was detected; IDH2 transcript expression in CD34+ cells derived from healthy donors and CML patients was assessed with Q-RT-PCR; the effects of IDH2 silencing on the growth and colony-forming cell (CFC) abilities of MEG-01 cells were studied. Results: iTRAQ analysis indicated that IDH2 expression was inhibited upon GAS2DN expression; western blot showed that GAS2DN inhibited IDH2 expression in both MEG-01 and BaF3/BCR-ABL cells, while GAS2 promoted its expression in BaF3/BCR-ABL cells, however IDH2 transcript expression didn\'t change; Calpain2 silencing partially "rescued" IDH2 inhibition caused by GAS2DN in K562 cells; the expression of IDH2 was significantly higher in CML CD34+ cells than control cells, and IDH2 silencing significantly inhibited the growth and the CFC abilities of MEG-01 cells. Conclusion: Targeting GAS2 inhibited the growth of CML cells partially through its down-regulation of IDH2.
