氨茶碱的免疫快速检测

摘要:通过制备氨茶碱的单克隆抗体,建立了氨茶碱的间接竞争酶联免疫吸附分析方法(ic-ELISA)。通过碳二亚胺法和活泼酯法将半抗原分别与牛血清蛋白(BSA)和卵清蛋白(OVA)偶联,获得免疫原和包被原。将免疫原免疫Balb/c小鼠,取其脾脏细胞与SP2/0 骨髓瘤细胞融合,得到产生阳性抗体的杂交瘤细胞株1G5。经过各项条件优化建立的间接竞争ELISA方法对氨茶碱的半抑制浓度为5.80 ng/ml,检测线性范围为1.62 ng/mL~20.66 ng/mL。抗体1G5与茶碱乙酸的交叉反应率小于20%。对添加到饲料基质中的氨茶碱进行检测, 结果回收率在94%~114%之间,变异系数小于10%,能用于实际样品中氨茶碱的检测。

Development of rapid immunoassay for aminophylline
Abstract:Monoclonal antibody against aminophylline was raised and used to develop an indirect competitive enzyme-linked immunoassay(ic-ELISA). The haptenactive conjugated to bovin serum albumin(BSA) and ovalbumin(OVA) as immounogen and coating antigen via 1-Ethyl-carbodiimide hydrochloride(EDC) and dicyclohexylcarbodiimide(DCC), respectively. One hybridoma-cell lines 1G5 secreting monoclonal antibodie against aminophylline was prepared by using mergenced myeloma cells SP2/0 of mouse with spleen cells from Balb/c mice immunized by the immounogen. The effect of several physicochemical factors on the monoclonal antibody for parathion was studied and the results were used to optimize the assay system. The IC50 was 5.80 ng/ml and the linear detection range (IC20~IC80) was 1.62 ng/mL~20.66 ng/mL measured by an indirect competitive ELISA. Cross-reactivities on acefylline was less 20%. The range of recovery rates of aminophylline from feed samples was 94%~114% and the variation coefficient was less than 10%. So monitoring aminophylline by using the monoclonal antibody is capable.
