新型硝基还原酶荧光探针的合成与应用

摘要:本文设计合成了一种新颖的检测硝基还原酶的双光子荧光探针。该探针是以香豆素奎唑啉酮为荧光团,对硝基苯为硝基还原酶的识别基团,在410 nm光激发下,该探针本身没有荧光,加入硝基还原酶后,探针在505 nm处有很强的荧光发射峰,呈现出"turn-on"型荧光探针。探针与硝基还原酶响应灵敏,检出限为11.54 ng/mL。此外,该探针具有好的选择性和生物相容性,并且成功用于活细胞内硝基还原酶双光子成像。

Synthesis and Application of a novel fluorescent probe for nitroreductase
Abstract:In this paper, a novel two-photon fluorescent probe is designed and synthesized for the detection of nitroreductase. The probe is designed by coumarin-quinazolinone as the fluorophore, p-nitrobenzyl moiety as the recognition group. Under the 410 nm light excitation, the probe itself has no fluorescence emission, but with the response to nitroreductase, the probe has an obvious fluorescence enhancement at 505 nm, which proved to be a "off-on" fluorescent probe. The probe is sensitive to nitroreductase and the detection limit is 11.54 ng/mL. In addition, the probe has good selectivity and biocompatibility, which was successfully used for two-photon fluorescence imaging of nitroreductase in living cells..
