摘要:目的:探究采用三维打印技术制备药物缓释支架及其用于乳腺癌治疗的可行性。方法:通过3D打印技术制备含有阿霉素(DOX)和顺铂(CDDP)两种药物的聚乳酸-羟基乙酸共聚物(PLGA)的载药支架,并采用SEM、MTT、流式分析、共聚焦成像及小动物多模式成像等方法证明载药支架的抗肿瘤效果。结果:SEM表征证明了载药支架与纯PLGA支架都具有完整的形态,药物被很好的包裹入支架中;MTT、流式分析及共聚焦实验证明载药支架相对于传统用药方式具有更好的促进肿瘤细胞凋亡的作用;利用小动物多模式成像系统验证了载药支架植入体内后的抗肿瘤效果,与小鼠肿瘤实物图共同表明其具有更好的抑制肿瘤生长的作用。结论:通过3D打印技术可以制备具有时间依赖性的DOX-CDDP-PLGA药物缓释支架,可以将其用作肿瘤局部治疗的新型药物递送系统,具有比较好的抗肿瘤效果。

3D printed drug-loaded stent for the treatment of breast cancer
Abstract:Objective: To investigate the tumor suppressive effect of 3D printed drug-loaded stent for breast cancer therapy. Methods: Stent containing doxorubicin (DOX), cisplatin (CDDP), and polylactic acid-glycolic acid copolymer (PLGA) were fabricated by 3D printing technology; SEM, MTT, flow cytometry assays, confocal experiment, and a small animal multi-mode imaging system methods were used to characterize the drug-loaded stent and its anti-tumor effect in vitro and in vivo. Results: SEM characterization confirmed that the drug-loaded stent and the pure PLGA stents have intact morphology, and drugs are well wrapped into the stents. MTT, flow cytometry and confocal experiments demonstrated that the drug-loaded stent has a better effect on promoting tumor cell apoptosis than the freely administered DOX, CDDP, DOX and CDDP combination drugs, the anti-tumor effect of drug-loaded stents was verified by small animal multi-mode imaging system and the mouse tumor physical map, indicating that it had a better inhibitory effect on tumor growth. Conclusion: Time-dependent DOX-CDDP-PLGA drug sustained-release stents fabricated by 3D printing technology had a relatively good anti-tumor effect as a novel drug delivery system for local treatment of tumors.
