薏苡花粉体外萌发及花药培养

摘要:为探索薏苡花粉体外萌发条件,建立薏苡花药培养及其愈伤组织增殖培养体系,本研究采用10种薏苡材料,通过观察不同时期不同薏苡花粉粒形态、测定花粉活力、确定花粉萌发条件、了解小孢子发育时期的形态特征,结果发现不同时期花粉粒大小不一,呈椭球形或球形,10种材料的花粉活力均在盛花期最大,广东、广西和河北薏苡花粉活力较大;四川白壳、蒲农一号和兴仁小白壳花粉活力较小。不同薏苡花粉体外萌发最适培养液不同,10种薏苡的最佳萌发条件是25℃培养 5~7 h。薏苡初花期花药发育是连续的,经单核期、双核期到三核期。小孢子处于单核靠边期时,花粉呈球形,仅有小液泡,该时期适合花药组织培养。不同品种其花药诱导率差异显著,兴仁小白壳、浙薏一号、广东薏苡、广西薏苡、福建薏苡、河北薏苡、辽宁薄壳在培养基MS+2,4-D1~2mg/L+KT1.5 mg/L成功诱导出愈伤组织,在分化培养基MS+IAA0.5mg/L+KT2 mg/L形成短小丛生芽。

In vitro Pollen Germination and Anther Culture of Coix lacryma-jobi L
Abstract:To explore job \'s-tears pollen germination in vitro condition, the establishment of job \'s-tears anther culture and callus multiplication culture system, this study adopts the 10 kinds of job \'s-tears material, by looking at different times different job \'s-tears pollen morphology, pollen vitality, pollen germination conditions were determined, period of microspore\'s shape characteristic, the results showed that the pollen grain sizes in different periods, the ellipsoid or globose, pollen vitality of 10 kinds of material in full bloom, the largest in guangdong, guangxi and hebei job \'s-tears pollen vitality;The pollen activity of sichuan white shell, punong no.1 and xingren small white shell was smaller.The optimum germination medium of pollen of coix was cultured at 25℃ for 5 ~ 7 h. The observation of the pollen grains of the early flowering stages showed that its development is continuous, with uninucleate, binucleate and trinucleate stages coexist in different ratios; the same piece of anther have microspores of different developmental stages. Microspores at the late uninucleate stage is suitable for anther tissue culture, the pollen is spherical with small vacuoles. During Coix anther culture, in the same culture medium, calluses and embryoids coexist. The anther\'s callus induction rate was in significant difference among different Coix cultivars. Calluses could be successfully induced from Xingrenxiaobaike, Zheyiyihao, Guangdong Coix, Guangxi Coix, Fujian Coix, Hebei Coix and Liaoning Boke Coix in the medium of MS+2, 4-D 1~2mg/L+KT1.5 mg/L. And Some Calluses of Coix could be successfully differentiated into short cluster buds in the medium of MS+IAA0.5mg/L+KT2 mg/L.
