抗病毒新药XYW小鼠体内药代动力学研究

摘要:目的:研究抗病毒新药XYW小鼠体内的药物代谢动力学特征。方法:应用高效液相色谱法建立并验证定量测定小鼠血浆中抗病毒新药XYW的方法。小鼠静注和肌注XYW后,测定血浆中不同时间点的血药浓度,应用Winnolin软件计算XYW药代动力学参数。结果:20mg/kg静注给药后CL为44.5±4.2mL/min/kg,V为35.6±11.6mL/kg。10、20和40 mg/kg三个不同剂量单次肌肉注射给药后,Tmax分别为4.2±1.56、4.4±1.96和4.4±1.96 min;Cmax分别为10.6±1.79、21.88±2.11和43.08±7.35 mg/L;AUC0-t分别为131.9±15.17、275.6±25.8和559.3±26.5 min mg/L。结论:高效液相色谱法定量测定小鼠血浆中抗病毒新药XYW的方法准确可靠。小鼠肌注不同剂量XYW后,Cmax、AUC分别与给药剂量呈良好的线性关系,平均生物利用度为61.1% 。

Study on pharmacokinetics of XYW in mice
Abstract:Objective: The aim of this experiment is to study the pharmacokinetics of XYW in mice. Methods: High performance liquid chromatography (HPLC) was used to establish and verify the quantitative determination of the antiviral drug XYW in mouse plasma. After mice were injected with XYW by intravenous injection or intramuscular injection, the blood drug concentration at different time points in plasma was determined, and the pharmacokinetic parameters of XYW were calculated by Winnolin software. Results: After intravenous injection of 20mg/kg, CL was 44.5± 4.2ml /min/kg and V was 35.6± 11.6ml /kg. After intramuscular injection of 10, 20 and 40 mg/kg, Tmax was 4.2±1.56, 4.4±1.96 and 4.4±1.96 min, respectively. Cmax was 10.6±1.79, 21.88±2.11 and 43.08±7.35 mg/L, respectively. The AUC0-t values were 131.9±15.17, 275.6±25.8 and 559.3±26.5 min mg/L, respectively. Conclusion: HPLC is an accurate and reliable method for the determination of XYW in mouse plasma. After intramuscular injection of XYW at different doses, Cmax and AUC showed a good linear relationship with the dose, and the average bioavailability was 61.1%.
