抗病毒新药XYW在大鼠体内的排泄研究

摘要:目的:本实验目的是研究XYW在大鼠体内的排泄情况。方法:大鼠肌肉注射XYW (20mg/kg),分别采用高效液相色谱(HPLC)法和同位素标记两种方法对大鼠尿、粪以及胆汁中XYW的原型药和代谢产物的排泄情况进行定量分析。结果:高效液相色谱法测定原型药:24小时原形药经粪便和尿排泄率分别为17.4%和5.53%。[3H]同位素标记法做物料平衡研究:48小时XYW及其代谢产物经粪便和尿排泄率分别为26.11%和63.19%;由尿及粪便累积总排泄率为给药总量的89.3%。另外,胆汁的排泄率为58%. 结论:结合色谱法与同位素标记法结果,可知原形药主要经粪排泄,而代谢产物主要经尿排泄。XYW在大鼠给药后48小时,该药排泄完全无体内蓄积现象。

Study on the excretion of XYW in Rats
Abstract:Objective: The aim of this experiment is to study the excretion profile of xyw in rats. Methods: A HPLC assay and [3H] isotope labelling method were developed and validated for the determination of xyw in rat urine, bile and feces. Results: After intramuscular injection of xyw(20mg/kg), it\'s prototype drug was determined by HPLC method: the excretion rate of xyw in feces and urine within 24 hours were 17.4% and 5.53% respectively. The mass balance study was measured by isotope labelling method: the excretion rate of xyw and it\'s metabolites in feces and urine within 48 hours were 26.11% and 63.19% respectively. The accumulative excretion rate in feces and urine totally was 89.3%;in addition,the excretion rate was 58% in rat bile. Conclusion: With the results of HPLC and isotope labelling methods, the data suggests that the prototype of xyw mainly excreted in feces, and the metabolites of xyw mainly excreted in urine.
