美国山核桃几个DNA片段的获得和验证

摘要:试图以美国山核桃叶片cDNA样品为模板获得目的片段,却偶然在实验中获得几个美国山核桃的DNA片段的序列。于是根据获得的序列设计了验证引物,重新提取叶片DNA,以此DNA为模板进行PCR,并对PCR产物测序,证明了这几个DNA序列真实存在。经过与GenBank中的相应数据比对分析可知,这几个DNA序列中,1个为基因组序列,1个为线粒体序列,2个为叶绿体序列。

Acquisition and verification of several DNA fragments of pecan
Abstract:An attempt was made to obtain a target fragment using a cDNA sample of pecan leaves as a template, but the sequence of several DNA fragments of pecan was obtained by chance in the experiment. Then, according to the obtained sequence, the verification primers were designed, the leaf DNA was re-extracted, the DNA was used as a template for PCR, and the PCR products were sequenced, which proved that these DNA sequences existed. After comparison with the corresponding data in GenBank, one of these DNA sequences is a genomic sequence, one is a mitochondrial sequence, and two are chloroplast sequences.
