P53在氡致人肺支气管上皮细胞(BEAS-2B)恶性转化中的作用

摘要:目的:研究P53基因在氡致人肺支气管上皮细胞(BEAS-2B)恶性转化中的作用。方法:将人肺支气管上皮细胞(BEAS-2B)的P53基因永久敲除,对P53基因敲除的BEAS-2B细胞和正常BEAS-2B细胞分别进行氡染毒处理,建立氡染毒致细胞恶性转化模型。使用WesternBloting、细胞增殖实验、软琼脂克隆实验、细胞侵袭实验和逆转录PCR实验等检测细胞恶性转化程度。结果:敲除P53基因后氡照射的BEAS-2B细胞更早地出现了恶性转化。相较于BEAS-2B细胞染氡组(2B-Rn),P53基因敲除的BEAS-2B细胞染氡组(P53 KO-Rn)增殖速度更快(P<0.05),形成克隆的时间更早(P<0.05),侵袭力更强(P<0.05),上皮-间质转化(EMT)相关基因mRNA表达水平变化也更早(P<0.05)。结论:氡照射导致人肺支气管上皮细胞(BEAS-2B)增殖加快,侵袭力增强,发生EMT改变,可以认为氡导致BEAS-2B细胞发生恶性转化,而P53基因是氡照射致BEAS-2B细胞恶性转化中的重要影响因素。

The role of P53 in radon-induced malignant transformation of BEAS-2B
Abstract:Objective: To investigate the role of P53 in radon-induced malignant transformation of human bronchial epithelial cells(BEAS-2B).Methods: The study permanently knocked out gene P53 in BEAS-2B,then the BEAS-2B and the P53 knockout BEAS-2B cells were treated by radon to establish the malignant transformed cell models.The malignant degree of cells was detected by western blotting, cell proliferation assay, soft agar cloning assay, cell invasion assay, reverse transcription PCR, etc.Results:After the P53 was knocked out,radon-irradiated BEAS-2B cells showed an earlier malignant transformation.Compared with the group of the BEAS-2B cells exposed to radon,the group of P53 knockout BEAS-2B cells exposed to radon had more powerful proliferation (P<0.05), Clones formed earlier(P<0.05), stronger invasive ability(P<0.05),and the mRNA expression level of EMT related genes changed earlier (P<0.05). Conclusion:Radon exposure led to accelerated proliferation,enhanced invasion and EMT changesof BEAS-2B, which could be considered as the malignant transformation of BEAS-2B cells caused by radon.P53 gene is an important factor influencing the malignant transformation of BEAS-2B cells induced by radon.
