尾孢菌与微小杆菌共培养促进尾孢菌素的生物合成

摘要:由于目前的化学合成或生物发酵法均难以满足尾孢菌素在光动力治疗和光催化等方面的应用需求,本研究尝试采用共培养发酵技术来提高尾孢菌素的产量。研究发现乙酰微小杆菌(Exiguobacteriumacetylicum Strain B001)与尾孢菌(Cercospora sp. JNU001)在共培养条件下可以明显促进尾孢菌素的生物合成,最高可达382mg/L,是原始菌株发酵培养条件的3倍。此外,显微镜形态观察和体外对抗实验(In vitro confrontation assay)揭示乙酰微小杆菌和尾孢菌之间通过一种全新的机制,即尾孢菌素嵌入和固定到乙酰微小杆菌的细胞膜上,以降低溶液中游离尾孢菌素来提高其产量。

Co-culture of Cercospora sp. JUN001 with E. acetylicum Strain B001 increases cercosporin proudction
Abstract:Owing to the limitation of cercosporin used in photodynamic theraphy and photocatalysis, which is currently produced by total chemical synthesis or microbial fermentation, co-cultivation method was here employed to enhance its production. We found that Exiguobacterium acetylicum Strain B001 isolated from microbial contanmination during preparation of cercosporin, dramatically increased cercosporin production when it was added to Cercospora sp. JNU001 culture with its final concentration of OD600=0.25 in S-7 medium. It reached to 382mg/L, with 3-fold increase. Importantly, a new mechanism that cercosporin secreted from Cercosporin sp. JNU001 inserted and accumulated into cell membrane of E. acetylicum to enhance its production was found, which was supported by in vitro confrontation assay and morphology of co-cultivation samples.
