急性脑卒中患者血清外泌体LncRNA表达谱差异性分析

摘要:目的:分析急性脑缺血患者血清外泌体LncRNA表达谱差异性,以期阐明脑缺血损伤的机理。方法:选择4例符合纳入标准的急性脑缺血患者和5例健康人,分离血清外泌体,运用生物芯片高通量检测LncRNA,同时对其进行GO、KEGG分析,筛选有差异表达的LncRNA。结果: 两组共有439个有统计学意义的差异基因,其中425个上调,14个下调;GO 富集分析前30个涉及生物过程的13个,细胞组分3个,分子功能14个;KEGG 富集分析,前30个涉及TGF-β、硒复合代谢、苯氨基丙酸代谢等通路。结论:急性脑缺血可导致血清外泌体LncRNA表达谱改变,涉及诸多通路和基因,可能参与了脑缺血损伤。

Analysis of the expression profile of exosomes lncRNA in patients serum with acute cerebral ischemia
Abstract:Objective: To analyzed the difference of lncRNA expression profile in serum exosomes of patients with acute cerebral ischemia, in order to elucidate the mechanism of cerebral ischemia injury. Motheds: Serum exosomes were isolated from 4 patients with acute cerebral ischemia and 5 healthy people, and lncRNA was detected by high-through put biochip, and the differential expression of lncRNA was screened by GO and KEGG analysis. Results: There were 439 statistic difference genes in the two groups, including 425 up-regulated genes and 14 down regulated genes. And according go analysis in Analysis of the expression profile of exosomes lncRNA in patients with acute cerebral ischemiahe first 30 of lncRNA involved 13 of biological processes, 3 of cell components and 14 of molecular functions; according KEGG enrichment analysis, the first 30 pathways includes TGF -β, seleniumcomplex metabolism, phenylaminopropionic acid metabolism and others. Conclusion: the expression profile of serum exosomes lncRNAs were changed after acute cerebral ischemia , that including many pathways and genes, these lncRNAs may be involved in cerebral ischemia injury.
