USP47对放射性皮肤损伤的影响及机制研究

摘要:目的:探究泛素特异性蛋白酶47(ubiquitin specific proteases 47,USP47)在放射性皮肤损伤中的作用和机制。方法:采用免疫荧光和Western blot检测人永生化表皮细胞(HaCaT)和人皮肤成纤维细胞(WS1)中USP47的表达和分布的情况;采用CCK8、EdU和划痕实验检测皮肤细胞的增殖迁移能力;采用流式细胞术检测细胞凋亡率;采用USP47基因敲除小鼠,建立小鼠放射性皮肤损伤模型,检测USP47对放射性皮肤损伤进展的影响;采用蛋白质组学对USP47基因敲除小鼠的受照皮肤进行检测,寻找差异蛋白。结果:受照后的皮肤HaCaT细胞与WS1细胞中USP47在核内表达升高;在HaCaT与WS1细胞敲低USP47后,细胞的增殖能力减弱,迁移能力减弱,细胞的凋亡率增加。动物水平上,USP47基因纯合缺失小鼠在受到电离辐射后皮肤损伤较野生型小鼠严重。采用蛋白质组学检测USP47敲除小鼠和野生型小鼠受照射皮肤组织中差异蛋白质,以差异表达量变化超过1.5作为变化阈值,共发现233个差异蛋白。结论:泛素特异性蛋白酶47(USP47)参与了放射性皮肤损伤的进展,并有望作为放射性皮肤损伤的防治新靶点。

Study on the effect and mechanism of USP47 on radiation skin injury
Abstract:Objective: To investigate the effect of ubiquitin-specific protease 47 (USP47) on radiation-induced skin injury and its mechanism. Methods: Immunofluorescence and Western Blot were used to detect the expression and distribution of USP47 in human immortalized epidermal cells (HaCaT) and human skin fibroblasts (WS1); CCK8, EDU and Scratch Tests were used to detect the proliferation and migration of skin cells ; Flow cytometry was used to detect the apoptotic rate; The model of radiation-induced skin damage was established by using USP47 knockout mice to detect the effect of USP47 on the progression of radiation-induced skin damage; Proteomics examined the irradiated skin of USP47 knockout mice to find differential proteins. Results: The expression of USP47 in the HaCaT and WS1 cells increased in the nucleusafter irradiation. When HaCaT and WS1 cells knocked down USP47, the cell\'s proliferation ability weakened, migration ability weakened, andcell apoptosis rate increased. At the animal level,mice with the USP47 gene knocked outhad more severe skin damage after exposure to ionizing radiation.Proteomics was used to detect differential proteins in the irradiated skin tissues of USP47 knockout mice and wild-type mice. With a differential expression of more than 1.5 as the change threshold, a total of 233 differential proteins were found.Conclusion: Ubiquitin-specific protease 47 (USP47) is involved in the progress of radiation-induced skin injury and is expected to serve as a novel target for the prevention and treatment of radiation-induced skin injury.
