煮沸法快速检测牛传染性鼻气管炎病毒

摘要:牛传染性鼻气管炎(IBR)是由牛传染性鼻气管炎病毒(IBRV)引起的一种呼吸道接触性传染病,在世界范围内流行。为了快速检测出IBRV,本研究提出一种简单有效的煮沸方法,不需要额外提取病毒DNA,通过实时荧光定量PCR检测手段,可以快速简便地检测出IBRV,节省时间降低成本。结果显示:通过高温煮沸,可以提取IBRV DNA,通过实时荧光定量PCR检测,不同批次间差异系数低于1%。本研究建立的牛传染性鼻气管炎病毒煮沸检测方法特异性好、可重复性强,可以用于早期IBRV的快速检测。

Rapid detection of infectious bovine rhinotracheitis virus using the boiling approach
Abstract:Bovine infectious rhinotracheitis (IBR) is a respiratory contact infectious disease caused by the bovine infectious rhinotracheitis virus (IBRV), which is endemic worldwide. In order to detect IBRV with high efficiency, our study proposes a simple and quicking boiling method that does not require additional extraction of viral DNA. IBRV can be quickly and easily detected and measured by fluorescent quantitative PCR detection, saving time and reducing costs. The results show that IBRV DNA can be extracted by boiling at high temperature, and the coefficient of variation within different batches is less than 1% by fluorescent quantitative PCR detection. The bovine infectious rhinotracheitis virus boiling detection method established in this study has good specificity and reproducibility, and can be used for the rapid and early detection of IBRV.
